摘 要:目的探讨跨膜4超家族成员1(TM4SF1)在肾细胞癌(RCC)中的表达及意义。方法应用实时荧光定量PCR和Western blot方法分析TM4SF1在肾癌A498细胞、os-rc-2细胞株中表达的改变,并与HK2人肾小管上皮细胞相比较,探讨TM4SF1在RCC发生发展中可能的作用。结果实时荧光定量PCR结果显示,与对照组相比较,TM4SF1基因在A498(t=20.24,P<0.01)和os-rc-2(t=23.93,P<0.01)肾癌细胞中mRNA表达水平均明显下降,差异具有统计学意义。Western blot方法检测显示,TM4SF1蛋白表达水平在A498(t=11.78,P<0.01)和os-rc-2(t=12.97,P<0.01)肾癌细胞中均显著低于对照组,差异亦具有统计学意义。结论TM4SF1在RCC中表达水平明显降低,说明其可能参与RCC发生发展的过程。Objective To investigate the expression and clinical significance of transmembrane-4-L-six-family-member-1(TM4SF1)in renal cell carcinoma(RCC).Methods RCC A498 cells and os-rc-2 cells were used as experimental group,Human renal tubular epithelial cells HK2 were used as matched group.The expression of TM4SFl mRNA and protein was examined by real-time quantitative PCR and Western blot.The difference in expression of TM4SFl between different groups was analyzed statistically.Results TM4SFl mRNA levels were lower in renal cancer A498 cells(t=20.24,P<0.01)and os-rc-2 cells(t=23.93,P<0.01)than in renal tubular epithelial cells HK2.There was significant difference in TM4SFl mRNA expression between renal tubular epithelial cells and renal cancer cells.TM4SF1 protein levels were also lower in renal cancer A498 cells(t=11.78,P<0.01)and os-rc-2 cells(t=12.97,P<0.01)than in renal tubular epithelial cells HK2.There was also significant difference in TM4SFl protein expression between renal tubular epithelial cells and renal cancer cells.Conclusions The gene and protein expression level of TM4SFl are dramatic declined in RCC,this suggests that TM4SFl may be related to the occurrence and development of RCC.
