三叶青提取物对口腔鳞状细胞癌生物学行为的体外研究

摘　　要：目的:研究三叶青提取物对口腔鳞癌细胞增殖、凋亡、侵袭、迁移的影响。方法:采用不同浓度的三叶青提取物处理人口腔鳞状细胞癌,检测CCK-8细胞增殖,Tunel试剂盒检测细胞凋亡,进行细胞迁移实验,Transwell试剂盒观察细胞侵袭能力,并用Western Blot检测相关蛋白的表达情况。结果:随着浓度的增加,三叶青提取物可以抑制癌细胞的增值及迁移,促进细胞凋亡,并促进Bax、cleave Caspase-3、cleave PARP蛋白的表达,降低Bcl-2、MMP-2、MMP-9的表达,差异均具有统计学意义(P<0.05)。结论:三叶青提取物能够有效抑制口腔鳞癌细胞增殖、侵袭和迁移,促进癌细胞凋亡。Objective:To study the effects of Trifolium repens extract on the proliferation,apoptosis,invasion and migration of oral squamous cell carcinoma cells.Methods:Different concentrations of Trifolium extract were used to treat human oral squamous cell carcinoma.CCK-8 was used to detect cell proliferation,Tunel kit to detect cell apoptosis,cell migration test,Transwell kit to observe cell invasiveness and Western Blot to detect the expression of related proteins.Results:With the increase of concentration,Trifolium repens extract can inhibit the proliferation and migration of cancer cells,promote cell apoptosis,and promote the expression of Bax,cleave Caspase-3,cleave PARP protein,reduce the expression of Bcl-2,MMP-2 and MMP-9,with statistical difference(P<0.05).Conclusion:Trifolium repens extract can effectively inhibit the proliferation,invasion and migration of oral squamous cell carcinoma cells,promote the apoptosis of cancer cells.
