摘  要:目的:分析20种茯苓菌株的亲缘关系。方法:应用简单重复序列间扩增(ISSR)标记分析20种茯苓菌株之间的遗传关系,并应用体细胞不亲和性实验对部分菌株之间的亲缘关系进行验证。结果:从128条引物中筛选出扩增图谱较好的20条引物进行ISSR分析。ISSR分析结果表明在遗传相似系数为36%时,L单独聚成一类,其余19种菌株聚成另一类;在遗传相似系数为94%时,除GIM 5.219与AS 5.137无法区分之外,其余菌株均可成功区分。选择遗传相似系数26%~94%中的9种菌株进行体细胞不亲和性分析,结果表明除GIM 5.219与L之间未产生拮抗线之外,其余测试菌株之间都产生较弱的拮抗线。结论:研究结果表明20条ISSR引物可以将20种菌株有效区分,除菌株L外,其他19种菌株遗传关系近,但体细胞不亲和性分析结果与ISSR分析结果不完全吻合。Objective:Genetic relationships of 20 Woifiporia Cocos strains were analyzed.Methods:Inter-simple sequence repeat(ISSR)marker was used to determine the genetic relationships of 20 W.cocos strains,and the results obtained from the ISSR marker analysis among the 9 W.cocos strains were validated by somatic incompatibility assays.Results:First,20 primers were screened from 128 primers and were subsequently used for ISSR-PCR analysis.And the ISSR analysis results showed that when the genetic similarity coefficient was set at 36%,strain L is separated from the other 19 strains,while the coefficient was set at 94%,except for GIM 5.219 and AS 5.137,the other strains are distinguished successfully.Second,somatic incompatibility assays were conducted among the selected 9 strains,which have the genetic similarity coefficient between 26%and 94%obtained from the ISSR analysis results.And the corresponding results indicated that except GIM 5.219 and L,weak barrage lines were generated among the tested 9 strains.Conclusion:Our results from this study suggested that 20 W.cocos strains were efficiently distinguished by ISSR marker analysis using the 20 primers,and that except strain L,the other 19 strians are genetically related.Moreover,the somatic incompatibility analysis results are not the same as the ISSR marker analysis results when both methods were used to determine the genetic relationships among the tested W.cocos strains.
