摘  要:目的应用Box-Behnken试验设计,优选超高压提取法提取金银花中绿原酸的最佳工艺。方法在单因素试验的基础上,采用3因素3水平Box-Behnken试验设计对金银花中绿原酸的超高压提取工艺参数进行优选。以绿原酸提取率为指标进行试验,考察液固比、提取压力、提取时间等因素对提取工艺的影响。结果超高压提取金银花中绿原酸的最佳工艺条件为液固比为75mL·g-1,提取压力100MPa,提取时间28s,此提取条件下绿原酸的提取率为24.68mg·g-1。结论超高压提取作为一种新兴的中药提取技术,具有提取率高、提取温度低、耗时短、能耗少等优点,为绿原酸提取提供一个全新的方法。OBJECTIVE To optimize the ultra-high pressure extraction(UPE) process of chlorogenic acid from Lonicera japonica by using Box-Behnken experimental design.METHODS On the basis of single factor screening,a three-factor and three-level Box-Behnken experimental design was employed,with liquid/material ratio(X1)、extracting pressure(X2),and extracting time(X3) as independent variables.The response variable was the extraction yield of chlorogenic acid.RESULTS The optimal conditions of UPE for chlorogenic acid were that the ratio of solvent to material(mL·g-1) was 75 mL·g-1,the extracting pressure was 100 MPa,and the extracting time was 28 second.With this extracting process,the extraction yield of chlorogenic acid was 24.68 mg·g-1.CONCLUSION As a novel extraction technology for Chinese herbal medicine,the UPE procedure has higher extraction yield,lower extracting temperature,shorter extacting time and less power consumption.The UPE has provided a brand-new method for extraction of chlorogenic acid from Lonicera japonica.
