摘　　要：目的探讨lnc RNA H19和SIRT6在胃癌中的表达及与预后的关系。方法选择收治的胃癌患者132例,实时荧光逆转录法及酶联免疫吸附法测定lnc RNA H19和SIRT6在胃癌组织及胃组织中的表达水平。采用Kaplan-Meier法估计不同lnc RNA H19和SIRT6表达水平的胃癌患者生存率,并采用Log-rank检验进行比较。结果胃癌组lnc RNA H19 mRNA表达水平高于癌旁组、SIRT6 mRNA表达水平低于癌旁组(P<0.05);lnc RNA H19、SIRT6呈黄色或深棕黄色颗粒,定于细胞核;胃癌组lnc RNA H19阳性102例、SIRT6阳性98例,胃癌组lnc RNA H19阳性率高于癌旁组,SIRT6阳性率低于癌旁组(P<0.05);lnc RNA H19、SIRT6蛋白表达与年龄无相关性(P>0.05);与淋巴血管间隙浸润、病理学分级、TNM分期、淋巴结转移、浸润深度、复发、肿瘤最大径有明显相关性,且有淋巴血管间隙浸润、病理学分期越高、TNM分期越高、有淋巴结转移、浸润深度越深、有复发、肿瘤最大径≥5 cm,lnc RNA H19蛋白阳性表达率越高,SIRT6蛋白阳性表达率越低(P<0.05);lnc RNA H19阴性组3年生存率及生存期均明显高于lnc RNA H19阳性组,SIRT6阳性组3年生存率及生存期均明显高于SIRT6阴性组(P<0.05)。结论胃癌患者中lnc RNA H19表达升高、SIRT6表达降低,与肿瘤增殖、浸润、迁移密切相关;lnc RNA H19低表达、SIRT6高表达的胃癌患者能获得较好的预后。Objective To investigate the expression of lnc RNA H19 and SIRT6 in gastric carcinoma tissues and its relationship with prognosis of patients.Methods The expression levels of lnc RNA H19 and SIRT6 in gastric cancer tissues and gastric tissues in 132 patients with gastric cancer were detected by real-time fluorescence reverse transcriptase and enzyme-linked immunosorbent assay(ELISA).Clinical and follow-up data of patients were collected.Kaplan-Meier method was used to estimate the survival rate of patients with gastric cancer with different expression levels of lnc RNA H19 and SIRT6,and Log-rank test was used to analyze and compare the survival rate in the patients with different expression levels of lnc RNA H19 and SIRT6.Results The expression levels of lnc RNA H19 mRNA in gastric cancer group were significantly higher than those in paratumor group(P<0.05),however,the expression levels of SIRT6 mRNA in gastric cancer group were significantly lower than those in paratumor group(P<0.05).The research results showed that the lnc RNA H19 and SIRT6 were yellow or dark brown-yellow particles,which located in the nucleus,and there were 102 cases of positive lnc RNA H19 and 98 cases of positive SIRT6 in gastric cancer group.The positive rate of lnc RNA H19 in gastric cancer group was significantly higher than that in paracancer group(P<0.05),however,the positive rate of SIRT6 in gastric cancer group was significantly lower than that in paracancer group(P<0.05).The expression levels of RNA H19 and SIRT6 protein were not correlated with patient’s age(P>0.05),however,which were positively correlated with lymphatic interstitial infiltration,pathological grade,TNM stage,lymph node metastasis,depth of invasion,recurrence and tumor maximal diameter(≥5cm)(P<0.05).Moreover the three-year survival rate and survival duration in lnc RNA H19 negative group were significantly higher than those in lnc RNA H19 positive group,and the three-year survival rate and survival duration in SIRT6 positive group were significantly higher than
