摘 要:目的:观察早期运动训练对急性脑梗死大鼠学习记忆能力及海马区神经细胞骨架相关蛋白表达的影响。方法:健康成年雄性Wistar大鼠60只,按随机数字表法分为假手术组、模型组、训练组,每组20只。采用线拴法建立大脑中动脉闭塞(MCAO)大鼠模型,术后24 h对大鼠进行强度渐增的跑台运动训练。于造模后14、28 d采用Morris水迷宫实验评估大鼠的空间学习记忆能力;干湿重法测定脑组织含水量;TTC染色检测大鼠脑梗死体积;免疫组织化学显色法检测海马CA1区微管蛋白β-tubulin的分布情况;免疫印迹检测海马组织中β-tubulin和微管相关蛋白-2(MAP-2)的表达变化;结果:与假手术组相比,模型组逃避潜伏期延长,穿越平台次数及平台象限停留时间均减少;脑含水量升高;海马组织β-tubulin与MAP-2蛋白表达量下调。与模型组比较,运动训练大鼠空间学习记忆能力显著提高,脑梗死体积和脑含水量明显降低,海马区β-tubulin与MAP-2蛋白水平显著升高。结论:大鼠脑梗死后早期进行运动训练可以减轻脑损伤程度,促进空间学习记忆能力的恢复,其机制可能与上调海马组织细胞骨架相关蛋白表达,增加神经可塑性相关。Objective:To investigate the effects of early exercise training on learning and memory function and expression of neuronal cytoskeletal proteins in the hippocampus of rats with acute cerebral infarction.Methods:Sixty male adult Wistar rats were divided randomly into sham group,model group and exercise group.The rats were subjected to left middle cerebral artery occlusion(MCAO)with the suture occlusion.They were then given exercise training with increasing intensity 24 h after MCAO.Morris water maze test was used to evaluate the spatial learning and memory ability 14 and 28 days after modeling.The volume of cerebral infarction in rats was detected by TTC staining.Immunohistochemistry was used to observe the distribution ofβ-tubulin in the CA1 region of the hippocampus.The expression ofβ-tubulin and microtube-associated protein-2(MAP-2)in the hippocampus was detected by Western blotting.Results:Compared with the sham group,the escape latency of rats in the model group was significantly longer,and the times of crossing the platform and the time in the platform quadrant were significantly reduced in the model group.Cerebral water content was significantly increased while the expression ofβ-tubulin and MAP-2 in hippocampal tissues was significantly decreased.Compared with the model group,the shorter latency and longer duration in Morris Water maze in the exercise group were observed,the cerebral infarction volume and cerebral water content were dramatically reduced,and the levels ofβ-tubulin and MAP-2 proteins in the hippocampus were remarkably increased.Conclusion:Early exercise training after MCAO reduces brain injury and improves spatial learning and memory,which may be associated with the upregulation in the expression of cytoskeletal proteins in the hippocampus and increased neuroplasticity in rats.
