摘 要:目的探讨玉米须多糖(Stigma Maydis polysaccharide,SMP)对高糖刺激下大鼠HBZY-1细胞增殖、炎症因子MCP-1、IL-6在基因及蛋白水平表达的影响.方法将实验细胞分为正常对照组(NG)、高糖组(HG)、高糖加不同浓度SMP组(SMP35组、SMP70组)4组,以35、70μg/mL两个质量浓度的SMP干预高糖刺激下的大鼠HBZY-1细胞,应用四甲基偶氮唑盐(MTT)光吸收法观察细胞增殖情况;应用RT-qPCR法检测HBZY-1细胞中MCP-1、IL-6 mRNA表达;应用Western Blot法检测HBZY-1细胞中MCP-1、IL-6蛋白表达.结果与NG组比较,HG组HBZY-1增殖加快(P<0.01),MCP-1、IL-6两指标的mRNA和蛋白表达均显著增加(P<0.01);与HG组比较,各质量浓度的SMP均可抑制高糖对大鼠HBZY-1细胞的增殖(P<0.01),mRNA和蛋白水平可下调MCP-1、IL-6的表达(P<0.01),且呈剂量依赖效应.结论SMP可能通过下调炎症因子MCP-1、IL-6表达,抑制高糖诱导的大鼠HBZY-1增殖,从而达到延缓糖尿病肾病发生、发展的作用.Objective To investigate the effects of Stigma maydis polysaccharide(SMP)on the proliferation of HBZY-1 cells stimulated by high glucose and the expression of monocyte chemoattractant protein-1(MCP-1)and interleukin-6(IL-6)at gene and protein levels.Method The experimental cells were divided into normal control group(NG),high glucose group(HG)and high glucose group with different concentration of SMP(SMP35 group and SMP70 group).The rats HBZY-1 cells were treated with SMP at 35 and 70μg/mL concentrations,the proliferation of rat HBZY-1 cells under high glucose stimulation was observed by the light absorption method of tetramethylazolium salt(MTT).The mRNA expression of MCP-1 and IL-6 in HBZY-1 cells was detected by RT-qPCR.The protein expression of MCP-1 and IL-6 in HBZY-1 cells was detected by Western Blot.Results Compared with the normal control group(NG group),HBZY-1 proliferation was accelerated in the high glucose group(HG group)(P<0.01),and mRNA and protein expressions of MCP-1 and IL-6 were significantly increased(P<0.01).Compared with HG group,the promotion effect of high glucose on the proliferation of rat HBZY-1 cells was inhibited in all SMP groups(P<0.01),and the expressions of MCP-1 and IL-6 were down-regulated at mRNA and protein levels(P<0.01),showing a dose-dependent effect.Conclusion SMP may play a role in delaying the occurrence and development of diabetic nephropathy by down-regulating the expression of inflammatory factors MCP-1 and IL-6 and inhibiting the proliferation of high-glucose-induced HBZY-1 cells in rats.
