摘 要:目的基于超高效液相色谱-高分辨飞行时间质谱联用技术(Ultra High performance Liquid Chromatography-High Resolution Time-of-flight Mass Spectrometry,UPLC-QTOF-MS/MS)对苦瓜醇提物中多肽的化学组成进行研究,建立快速准确的分析方法。方法采用Welch Ultimate UPLC XB-C18色谱柱(100mm×2.1mm, 1.8μm),以0.1%TFA水溶液-乙腈为流动相以0.3ml/min流速梯度洗脱,电喷雾源(ESI)、QTOF作检测器,正离子模式检测。质谱产生的原始数据采用Mascot软件进行分析。结果通过检索Momordica-charantia.fasta数据库,共检测出了来自苦瓜的62种蛋白质的86种肽段。结论建立了苦瓜醇提物中多肽类成分的快速检测方法,为其质量控制提供参考。Objective To establish a rapid and accurate method for the determination of polypeptide in Momordica charantia L. alcohol extract based on Ultra High performance Liquid Chromatography-High Resolution Time-of-flight Mass Spectrometry(UPLC-QTOF-MS). Methods The analysis was performed by Welch Ultimate UPLC XB-C18 column(100 mm×2.1 mm, 1.8μm) with the mixture of 0.1%TFA aqueous solution-acetonitrile as mobile phase by gradient elution, and the flow rate was 0.3 ml/min. QTOF equipped with ESI ion source was performed in positive ionization, finally, Mascot software was used to analyze the original data generated by mass spectrometry.Results A total of 86 peptide segments of 62 proteins from balsam pear were detected by searching Momordica charantia.fasta database. Conclusion A rapid method for the determination of polypeptides in Momordica charantia L. alcohol extract was established to provide reference for its quality control.
