摘 要:目的探讨非小细胞肺癌患者丝氨酸羟甲基转移酶(SHMT2)与CpG岛甲基化的相关性。方法选取我院2015年5月至2018年3月期间非小细胞肺癌患者90例,病理癌组织纳入观察组,癌旁(距病灶2 cm)组织纳入对照组。采用酶联免疫吸附试验(ELISA)双抗夹心法测定SHMT2表达水平;采用CIMP评估CPG岛甲基化率。结果癌组织SHMT2水平显著高于癌旁组织SHMT2,差异有统计学意义(P<0.01)。癌组织CpG岛甲基化率CIMP水平显著高于癌旁组织,差异有统计学意义(P<0.05)。SHMT2在Ⅰ/Ⅱ期的表达水平显著低于Ⅲ/Ⅳ期,差异有统计学意义(P<0.01)。SHMT2在N0期的表达水平显著低于N1/N2/N3期,差异有统计学意义(P<0.01)。结论非小细胞肺癌SHMT2表达高于癌旁组织;非小细胞肺癌CpG岛异常甲基化高于癌旁组织;CpG岛异常甲基化与SHMT2存在显著的相关性(P<0.01),非小细胞肺癌患者辅助去甲基化治疗可能会提高患者治疗效果。Objective To investigate the correlation between serine hydroxymethyl transferase(SHMT2)and CpG island methylation in patients with non-small cell lung cancer(NSCLC).Methods Included were 90 NSCLC patients in our hospital between May 2015 and March 2018.From these patients,cancerous tissue samples were assigned to the study group,and cancer-adjacent tissue samples(2 cm from the lesion)were assigned to the control group.The expression level of SHMT2 was determined by double-antibody sandwich ELISA;the methylation of CPG island was evaluated by CpG island methylator phenotype(CIMP).Results The expression level of SHMT2 in cancer tissue was significantly higher than that in cancer-adjacent tissue,with statistically significant difference(P<0.01).The CpG island methylation rate as measured by CIMP level in cancer tissue was significantly higher than that in cancer-adjacent tissues,with statistically significant difference(P<0.01).The SHMT2 expression level in stageⅠ/Ⅱcancers was significantly lower than that in stageⅢ/Ⅳcancers,with statistically significant difference(P<0.01).The SHMT2 expression level in stage N0 cancers was significantly lower than that in stage N1/N2/N3 cancers,with statistically significant difference(P<0.01).Conclusion In NSCLC,SHMT2 expression level in cancers is higher than that in cancer-adjacent tissues,and so is the aberrant CpG island methylation.There is a significant correlation between aberrant CpG island methylation and SHMT2.Addition of demethylation therapy for NSCLC patients may help improve treatment efficacy.
