摘 要:目的:分离培养泰安市奈河水系城区段水样中具有硝化能力的细菌,并对其进行分离培养和鉴定。方法:采集泰安市奈河水系城区段的水样,使用选择性培养基进行增菌培养、分离培养和纯培养,获得纯培养的菌株并保存;对水样中分离培养得到的细菌进行硝化能力检测、16SrDNA的基因测序与对比、形态特征观察、生化实验与鉴定。结果:分离一株具有硝化能力的菌株,测序分析该菌株属于黄杆菌属,生化反应鉴定该株细菌为金黄杆菌Ⅱb。结论:泰安市奈河水系城区段水样中含有异养型硝化细菌,其在生态循环中可能发挥重要作用。Objective:to isolate,culture and identify the bacteria with nitrification ability in the water samples of Naihe City,Taian City.Methods:water sample was collected from the urban area of Naihe river system in Taian City.The pure culture strains were obtained and preserved by enrichment culture,isolation culture and pure culture.The isolated bacteria were tested for nitrification ability.The strains with nitrification ability were sequenced and compared for 16SrDNA,morphological observation and biochemical identification.Results:a strain with nitrification ability was isolated.Sequencing showed that it belonged to Flavobacterium sp.,and biochemical identification showed that it belonged to chrysobacteriumⅡB.Conclusion:heterotrophic nitrifying bacteria may play an important role in the ecological cycle.
