摘  要:目的:探讨小檗碱(BBR)对博来霉素(BLM)诱导的大鼠肺纤维化干预作用及机制。方法:将24只健康雄性SD大鼠随机分为Sham组、BLM组、BBR组和BLM+BBR组,每组6只。BLM组和BLM+BBR组气管内滴注BLM(5 mg·kg^-1,0.2 mL)复制肺纤维化大鼠模型。BBR组和BLM+BBR组自术后第2天起,连续行BBR(200 mg·kg^-1·d^-1)灌胃28 d,余组给予同体积生理盐水灌胃,各组均于第28天取材。观察肺系数、肺组织病理变化,ELISA法检测大鼠肺组织Ⅰ型胶原(COLⅠ)和Ⅲ型胶原(COLⅢ)含量,免疫组化检测各组肺组织TGF-β1的表达。结果:BLM组大鼠肺组织结构紊乱、炎症浸润伴胶原含量增加,而BLM+BBR组肺组织结构损伤程度较BLM组轻。BLM组肺系数[(7.60±1.03) mg·g^-1]高于Sham组[(5.66±1.09) mg·g^-1](P<0.05),BLM+BBR组肺系数[(5.86±1.08) mg·g^-1]低于BLM组(P<0.05)。ELISA结果显示,BLM组肺组织COLⅠ[(3.46±0.24)vs(3.08±0.21) ng·mL^-1]、COLⅢ含量[(0.19±0.02)vs(0.13±0.02) ng·mL^-1]高于Sham组(P<0.05),BLM+BBR组肺组织COLⅠ(3.11±0.20) ng·mL^-1、COLⅢ(0.15±0.03) ng·mL^-1含量低于BLM组(P<0.05)。免疫组化检测结果显示BLM组TGF-β1的平均光密度[(0.64±0.16)vs(0.48±0.17)]高于Sham组(P<0.05),BLM+BBR组TGF-β1平均光密度(0.49±0.13)低于BLM组(P<0.05)。结论:小檗碱可降低肺纤维化大鼠肺组织TGF-β1表达及胶原含量,从而改善大鼠肺纤维化程度。Objective: To explore the effect and mechanism of berberine(BBR) on bleomycin(BLM)-induced pulmonary fibrosis in rats. Methods: 24 healthy male SD rats were randomly divided into Sham group, BLM group, BBR group and BLM+BBR group with 6 rats in each group.The pulmonary fibrosis model was induced by intratracheally injection with bleomycin(5 mg/kg,0.2 ml)in BLM group and BLM+BBR group. The rats in BBR group and BLM+BBR group were fed Ber 200 mg·kg^-1·d^-1 BBR from the second day,others groups were fed the same volume physiological saline solution for 28 days. At the end of the 28 th day, lung coefficient were measured and pathological changes of lungs tissue were observed by light microscopy. Contents of COLI and COLIII in lung tissues were measured by ELisa. The expression of TGF-β1 was detected by immunohistochemical method. Results: The lung tissue structure of BLM group was disordered and the inflammatory infiltration was accompanied by the increase of collagen content, while the lung tissue structure of BLM+BBR group was less damaged than that of BLM group. The lung coefficient of the BLM group [(7.60±1.03) mg·g^-1] was higher than that of the Sham group [(5.66±1.09) mg·g^-1](P<0.05), and it lower in BLM+BBR group[(5.86±1.08) mg·g^-1] than in BLM group(P<0.05).ELISA results showed the levels of COL Ⅰ [(3.46±0.24) ng·mL^-1 and COL Ⅲ[(0.19±0.02) ng·mL^-1] in the lung tissues of the BLM group were higher than those in Sham group [(3.08±0.21) ng·mL^-1,(0.13±0.02) ng·mL^-1], and the levels of COL Ⅰ[(3.11±0.20) ng·mL^-1 ]and COL Ⅲ[(0.15±0.03) ng·mL^-1 ] in the BLM+BBR group decreased significantly compared with BLM group(P<0.05). The results of immunohistochemistry showed that the average optical density of TGF-β1 in BLM group [(0.64±0.16) vs(0.48±0.17)] was higher than that in Sham group(P<0.05), and the average optical density of TGF-β1 in BLM+BBR group(0.49±0.13) lower than BLM group(P<0.05).
