摘  要:目的观察伊沙佐米对人胃癌细胞株SGC-7901生物学行为的影响,并探讨其可能的机制。方法取对数生长期、状态良好的SGC-7901细胞,随机分为12.5μmol/L组、25μmol/L组、50μmol/L组、100μmol/L组和对照组,分别加入终浓度为12.5、25、50、100μmol/L的伊沙佐米及相同体积PBS。各组分别于作用24、48、72 h,采用CCK-8法检测细胞增殖能力(光密度值)。各组作用24 h,采用流式细胞术检测细胞凋亡率,Transwell迁移、侵袭实验计数穿膜细胞数,Western blotting法检测Wnt、β-catenin、NF-κB蛋白表达。结果随着时间的延长,各组细胞增殖能力均呈升高趋势。相同作用时间下,对照组、12.5μmol/L组、25μmol/L组、50μmol/L组、100μmol/L组细胞增殖能力依次降低,两组间比较P均<0.05。对照组、12.5μmol/L组、25μmol/L组、50μmol/L组、100μmol/L组细胞凋亡率依次升高,迁移和侵袭实验穿膜细胞数及Wnt、β-catenin、NF-κB蛋白相对表达量依次降低,两组间比较P均<0.05。结论伊沙佐米可抑制SGC-7901细胞增殖、迁移及侵袭,并促进细胞凋亡,其机制可能与抑制Wnt/NF-κB信号通路有关。Objective To explore the inhibitory effect of ixazomib on human gastric cancer cell Line SGC-7901 and its mechanism.Methods SGC-7901 in the logarithmic phase were divided into five groups,namely 12.5,25,50,and 100μmol/L groups and the control group,which were added with 12.5,25,50,and 100μmol/L ixazomib and the same volume of PBS.CCK-8,flow cytometry,and Transwell assays were used to analyze cell proliferation,apoptosis,migration and invasion at 24,48,and 72 h.Western blotting was used to detect the expression of Wnt,β-catenin and NF-κB protein.Results All groups showed increased cell proliferation over time.Compared with the control group,the cell proliferation abilities of the 12.5,25,50 and 100μmol/L groups decreased significantly(all P<0.05).In the control group,12.5,25,50,and 100μmol/L groups,the apoptotic rates increased in sequence,and the migration and invasion experiments showed that the number of transmembrane cells,and the relative protein expression of Wnt,β-catenin,and NF-κB decreased successively,with statistically significant difference between every two groups(all P<0.05).Conclusion Ixazomib can inhibit the cell proliferation,migration and invasion,but promot the apoptosis by inhibiting the Wnt/NF-κB signaling pathway.
