半夏泻心汤含药血清对HP感染GES-1细胞DNA甲基化CpG结合域蛋白与甲基转移酶 mRNA表达的影响

摘　　要：目的探讨半夏泻心汤对幽门螺杆菌(Helicobacter pylori,HP)感染GES-1细胞DNA甲基化CpG结合域蛋白与甲基转移酶mRNA表达的影响。方法临床收集HP相关消化性溃疡(Peptic ulcer,PU)患者中药组半夏泻心汤治疗前后及西药组治疗后含药血清。将上述血清分别作用于经HP感染的GES-1细胞用药前组、中药组、西药组,另设对照组。干预24 h后收集细胞,实时荧光定量PCR法检测细胞MECP2、MBD1、DNMT3A、DNMT3B mRNA表达水平。结果用药前组MECP2 mRNA与对照组、中药组比较有显著性差异(P<0.01,P<0.05);对照组与中药组之间没有显著性差异。MBD1、DNMT3A、DNMT3B mRNA各组之间没有显著性差异。结论半夏泻心汤可通过下调或抑制MECP2 mRNA表达,从而参与细胞DNA甲基化调控,这可能是其治疗HP相关PU的作用机制之一。Objective To explore the mRNA expressions of DNA methyl-CpG-bindingdomain(MBD)protein and methyltransferase(DNMT)in GES-1 cells infected by Helicobacter pylori(HP)treated with human medicated serum of Banxia Xiexin Decoction(BXD).Methods The peptic ulcer(PU)patients were randomly assigned into Chinese or western medicine treatment group.We collected the medicated serum after treatment with BXD or conventional western medicine for 7 days.After infected by HP,the serum above was added to the GES-1 cell groups of Chinese medicine and western medicine,respectively.Meanwhile,we set up the control groupand the prior treatment group.24 h later,cells were collected.The mRNA expressions of MECP2,MBD1,DNMT3 A and DNMT3 B were detected by real-time quantitative PCR(RT-qPCR).Results About the expression of MECP2 of the prior treatment group,there were significant differences compared with the control andChinese medicine group(P<0.01,P<0.05).There was no difference between the latter two groups.There was no significant difference among the groups in the mRNA expressions of MBD1,DNMT3 A and DNMT3 B.Conclusion BXD can suppress or down-regulate the expression of MECP2 in GES-1 cells infected by HP,which might be one of its mechanisms in treating HP-related PU.
