摘 要:目的:研究MicroRNA338(MiR-338)和多效蛋白(PTN)在卵巢良性疾病组织、交界性病变组织及上皮性卵巢癌(Epithelial ovarian cancer,EOC)中表达及其对EOC的诊断价值。方法:通过免疫组织化学检测70例卵巢良性病变组织、80例卵巢交界性病变组织和70例卵巢癌组织中MiR-338及PTN的表达,同时酶联免疫吸附试验(ELISA)检测血清MiR-338及PTN表达水平,并对MiR-338及PTN的表达与相关临床病理情况进行分析。结果:MiR-338及PTN在各类型的良性病变组织中几乎不表达,卵巢癌中MiR-338及PTN表达阳性率及其评分分别为62.0%,5.0±1.2;68.5%,5.7±1.1,明显高于卵巢良性疾病(4.3%,0.6±0.2;7.1%,0.8±0.2)及卵巢交界性病变组织(8.6%,1.1±0.4;10.0%,1.3±0.3),结果具有高度显著差异(P<0.05)。不同临床分期卵巢癌患者MiR-338及PTN浓度与TNM分期之间经方差分析,显示各项指标之间均有显著性差异(P<0.05),且MiR-338表达水平与PTN呈正相关。结论:MiR-338及PTN在EOC组织中高表达,PTN具有促血管新生、刺激细胞增殖与迁移等多种生物学活性,MiR-338对PTN导致的卵巢癌淋巴结转移具有间接促进作用,MiR-338及PTN表达可能是反映EOC生物学行为,MiR-338及PTN的检测有助于判断有无淋巴结转移及预后。因此,联合MiR-338及PTN检测对于卵巢癌的诊断具有重要价值。Objective:To study the expression of MicroRNA338(MiR-338)and(PTN)in benign ovarian diseases,borderline lesions and epithelial ovarian cancer(Epithelial ovarian cancer,EOC)and their diagnostic value in EOC.Methods:The expression of MiR-338 and PTN and related clinicopathological conditions in 70 cases of benign ovarian lesions,80 cases of borderline ovarian lesions and 70 cases of ovarian cancer were detected by immunohistochemistry.At the same time,the levels of serum MiR-338 and PTN were detected by enzyme-linked immunosorbent assay(ELISA).Fuethermore,the expression of MiR-338 and PTN and related clinicopathological conditions were analyzed.Results:There was almost no expression of MiR-338 and PTN in various types of benign lesions.The positive rate of MiR-338 and PTN expression and their scores in ovarian cancer were 62.0%,5.0±1.2;68.5%,5.7±1.1 respectively,which were significantly higher than that in benign ovarian diseases with the expression and their scores 4.3%,0.6±0.2;7.1%,0.8±0.2 respectively and borderline lesions with the expression and their scores 8.6%,1.1±0.4;10.0%,1.3±0.3 respectively.And there was significantly higher in ovarian cancer than that in benign ovarian diseases(P<0.05).The analysis of variance between the concentrations of MiR-338 and PTN in patients with ovarian cancer of different clinical stages and TNM stages showed that there were significant differences among all indexes(P<0.05),and the expression levels of MiR-338 and PTN were positively correlated.Conclusion:There were high expression of MiR-338 and PTN in malignant ovarian cancer tissues.PTN has many biological activities such as promoting angiogenesis,stimulating cell proliferation and migration.MIR-338 can indirectly promote lymphnode metastasis of ovarian cancer induced by PTN.The expression of MiR-338 and PTN may reflect the biological behavior of ovarian cancer.MiR-338 and PTN are benificial to dignosis whether there is lymph node metastasis and prognosis or not.Therefore,the combined detection of MiR-338 and PTN is
