摘 要:目的探讨人参茎叶总皂苷对大鼠MNU所致RP的作用及机制。方法随机将大鼠分成空白对照组、MNU组(模型组)和人参茎叶总皂苷组(治疗组),模型组和治疗组分别腹腔注射注射(ip)MNU 60 mg/kg复制RP模型,对照组则ip等量生理盐水,治疗组连续7 d灌胃人参茎叶总皂苷50 mg/kg。进行视觉电生理检测,视网膜的病理组织学,采用Morris水迷宫检测视觉功能,采用流式细胞仪检测Beclin-1和LC3蛋白的表达。结果治疗组与模型组比较,视觉电生理检测、病理组织学检查显示,模型组视网膜电图波形振幅基本消失,动物视网膜外核层受损,细胞稀疏;人参茎叶总皂苷明显减轻上述的症状;水迷宫实验显示,治疗组与模型组比较总时间、潜伏期、路程、速度低于模型组;治疗组与模型组比较,Beclin-1和LC3表达降低。结论人参茎叶总皂苷对大鼠MNU所致RP具有保护作用,可能与降低Beclin-1和LC3表达有关。Objective To investigate effect of ginsenosides on retinal pigmentosa induced by MNU in rats.Methods The rats were randomly divided into control group,MNU group(model group)and Ginsenosides group(treatment group).The model group and treatment group were intraperitoneally injected with 60 mg/kg MNU to replicate the RP model,while the control group received IP equal amount of normal saline,and the treatment group were given 50 mg/kg Ginsenosides for 7 consecutive days.Visual electrophysiological examination was performed,and retinal histopathology was performed;visual function was detected by Morris water maze,and beclin-1 and LC3 protein expression was detected by flow cytometry.Results Compared with the model group,visual electrophysiological examination and histopathological examination showed that the waveform amplitude of the electroretinogram in the model group basically disappeared,the outer nucleus layer of the retina was damaged,and the cells were sparse.Ginsenosides significantly alleviated these symptoms.The water maze experiment showed that the total time,incubation period,distance and speed of the treatment group were lower than those of the model group.Beclin-1 and LC3 expression was decreased in the treatment group compared with the model group.Conclusion Ginsenosides has protective effect on RP induced by MNU in rats,which may be related to the decreased beclin-1 and LC3 expression.
