摘  要:目的研究细粒棘球绦虫原头蚴(EgPSCs)对肝癌HepG2细胞增殖和侵袭的影响。方法加入不同数量EgPSCs(0、50、100和200个/孔)与肝癌细胞HepG2共培养,采用MTT、细胞划痕实验和transwell检测,分别检测不同时间点,各实验组HepG2细胞的增殖、迁移与侵袭能力。结果MTT结果表明,50、100和200个/孔的EgPSCs48、72、96 h的增殖能力均显著高于0个/孔的EgPSCs(P<0.05);细胞划痕实验结果表明,培养24、48 h时100、200个/孔的EgPSCs比0个/孔EgPSCs的剩余划痕面积显著减小(P<0.05);transwell结果表明,100个和200个/孔EgPSCs实验组的侵袭细胞数显著高于0个/孔EgPSCs实验组(P<0.01)。结论体外研究表明,EgPSCs和肝癌HepG2细胞共培养,能促进肝癌细胞的增殖、迁移、侵袭,为明确细粒棘球绦虫对肝癌的分子作用机制奠定基础。Objective The effect of the protoscoleces of Echinococcus granulosus(EgPSCs)on the proliferation of hepatocellular carcinoma cell is not clear.This study is to explore the effect of EgPSCs on the proliferation and invasion of hepatocellular carcinoma at the cellular level.Methods HepG2 cells were co-cultured with EgPSCs(0,50,100 and 200/well).MTT,cell scratch assay and transwell assay were used to detect the proliferation,migration and invasion of HepG2 cells at different time points.Results MTT results showed that the proliferation of(50,100 and 200/well)EgPSCs at 48,72 and 96h were significantly higher than that of 0/well EgPSCs(P<0.05).Cell scratch test showed that wound remaining of(100 and 200/well)EgPSCs were significantly smaller than that of 0/well EgPSCs at 24 and 48 h(P<0.05);Transwell results showed that the number of invasive cells in 100 and 200/well EgPSCs group were significantly higher than that in 0/well EgPSCs group(P<0.01).Conclusion Co-culture of EgPSCs and HepG2 cells can promote the proliferation,migration and invasion of hepatocellular carcinoma cell,and lay a foundation for the molecular mechanism of E.granulosus on hepatocellular carcinoma.
