摘 要:目的检测逆转座子LINE-1在脑胶质瘤中的表达水平,探讨其异常激活与肿瘤病理特征和患者生存预后的关联;方法收集30例脑胶质瘤组织,利用qPCR检测逆转座子LINE-1ORF1的表达水平,并分析LINE-1表达水平与肿瘤病理分类的关系,利用Kaplan-Meirer生存曲线分析LINE-1异常活化与患者生存预后的关系;结果与癌旁组织相比,脑胶质瘤组织中LINE-1的表达水平显著升高,同时在WHO分级Ⅲ-Ⅳ级肿瘤组织LINE-1的表达量明显高于Ⅰ-Ⅱ级,Ki-67高表达的脑胶质瘤组织LINE-1也有着更高的表达(P<0.05),与LINE-1低表达的脑胶质瘤患者相比,LINE-1高表达的脑胶质瘤患者生存期显著缩短。结论逆转座子LINE-1在脑胶质瘤中存在异常活化,有潜力成为不良预后的分子标志物。Objective To detect the expression level of retrotransposon in glioma and analyze its effect on clinicopathological features and survival prognosis.Methods 30 Patients with glioma undergoing surgical were selected.Glioma tissues and precancerous tissue were collected.qPCR was used to detect the expression of LINE-1 in glioma tissue.The overall survival time of glioma patients were followed up.Results Compared with paraneoplastic tissues,the expression of LINE-1 in glioma tissues was significantly increased.The expression ofLINE-1 in glioma with WHO gradeⅢ-Ⅳand high Ki-67 expression were significantly higher than those in glioma withⅠ-Ⅱgrade and low expression of Ki-67(P<0.05).Compared with glioma patients with low expression of LINE-1 free survival time of glioma patients with high expression ofLINE-1 were significantly shortened.Conclusion The retrotransposon LINE-1 has showed abnormal activation in glioma,which has the potential to be a molecular marker of poor prognosis.
