摘 要:目的探讨HPV型别与SOX1和PAX1基因甲基化的相关性。方法共收集广州中医药大学深圳医院(福田)322例宫颈脱落细胞样本,分别应用导流杂交技术和荧光定量PCR检测HPV型别和甲基化水平,比较SOX1和PAX1基因在不同HPV型别中的甲基化水平,以及感染不同风险HPV型别的患者在不同级别宫颈病变中的甲基化检出率。结果在感染最高风险致癌型别HPV16/18患者中,SOX1和PAX1基因甲基化检出率最高为54.9%;其次是感染非16/18hrHPV患者中SOX1和PAX1基因甲基化检出率为24.0%;在感染其他型别HPV患者中SOX1和PAX1基因甲基化检出率为11.1%;在HPV检测阴性患者中SOX1和PAX1基因甲基化检出率为2.2%。无论在HPV16/18阳性、非HPV16/18hrHPV阳性或者其他型别HPV阳性下的不同级别宫颈病变者的甲基化检出率都是随病变级别的加深而增大。结论SOX1和PAX1基因甲基化检出率与感染的HPV型别有关,感染hrHPV的患者其SOX1和PAX1基因甲基化水平高于感染其他HPV型别的患者。Objective The aim was to investigate the correlation between HPV type and methylation of SOX1 and PAX1 genes.Methods A total of 322 cervical exfoliated cell samples were collected from Shenzhen Hospital(Futian)of Guangzhou University of Chinese Medicine,and HPV type and methylation levels were detected by flow-through hybridization and quantitative PCR,respectively.The methylation levels of SOX1 and PAX1 genes in different HPV types were compared.And the detection rate of methylation were compared in patients with different types of HPV types at different levels of cervical lesions.Results Among patients with the highest risk of carcinogenic HPV16/18 infection,the highest detection rate of SOX1 and PAX1 gene methylation was 54.9%;secondly,the detection rate of SOX1 and PAX1 gene methylation was 24.0 among patients with non-16/18 hrHPV infection;The detection rate of SOX1 and PAX1 gene methylation was 11.1%in patients infected with other types of HPV;the detection rate of SOX1 and PAX1 gene methylation was 2.2%in HPV negative patients.Regardless of HPV16/18 positive,non-HPV16/18 hrHPV positive or other types of HPV positive,the methylation detection rate of patients increased with the severity of cervical lesions.Conclusion The detection rate of SOX1 and PAX1 gene methylation is related to the infected HPV type.Patients infected with hrHPV had higher levels of methylation of SOX1 and PAX1 genes than patients infected with other HPV types.
