摘　　要：目的探讨Survivin及PTEN蛋白在胃癌组织中的表达特点,分析其表达与临床病理因素及预后的关系。方法收集经病理证实的有完整病历资料的胃癌300例石蜡组织标本为胃癌组,相应癌旁正常组织120例作为对照组。应用免疫组织化学HP法对切片进行检测。结果Survivin在胃癌组和对照组中表达率分别为72.0%和5.0%,而PTEN表达率分别为34.0%和92.5%,差异均有统计学意义(P<0.05);两者阳性表达率与组织学类型、分化程度、浸润深度、远处转移及病理分期等有关(P<0.05);Survivin表达与PTEN表达间存在负相关性(P<0.05,γ=-0.524)。两者表达水平与生存率有关(P=0.000,P=0.000),Survivin表达的增强,PTEN表达的减弱与不良预后有关。结论胃癌中Survivin表达上调及PTEN表达下调与胃癌发生、转移及预后密切相关。两者可能在胃癌的侵袭、转移中存在共同作用,相互影响,可能为候选治疗靶点。Objective To investigate the expression of Survivin and PTEN proteins in gastric cancer tissues,and to analyze the relationship between the expression and clinicopathological factors and prognosis.Methods 300 cases of paraffin-embedded tissues of gastric cancer with pathologically confirmed complete medical records were collected as gastric cancer group,and 120 cases of normal adjacent tissues were used as control group.Using immunohistochemical HP and analyzed using statistical software.Results The expression rates of Survivin in gastric cancer group and control group were 72.0%and 5.0%,respectively,while the PTEN expression rates were 34.0%and 92.5%,respectively.The difference was statistically significant(P<0.05).The positive expression rate of the two groups were related to histological type,degree of differentiation,depth of invasion,distant metastasis and pathological stage(P<0.05),there was a negative correlation between Survivin and PTEN expression(P<0.05,γ=-0.524).The expression levels of the 2 were related to survival(P=0.000,P=0.000),the expression of Survivin was enhanced,and the decrease of PTEN expression was associated with poor prognosis.Conclusion Up-regulation of Survivin expression and down-regulation of PTEN expression in gastric cancer were closely related to the occurrence,metastasis and prognosis of gastric cancer.The 2 may have a synergistic effect on the invasion and metastasis of gastric cancer,which may be candidate therapeutic targets.
