摘　　要：目的探讨增生型糖尿病视网膜病变(PDR)患眼玻璃体、增生膜中长链非编码RNA浆细胞瘤变异易位基因1(lncRNA PVT1)、微小RNA-26b(miR-26b)表达水平及意义。方法选取2016年3月至2019年3月本院收治的单眼发病的糖尿病视网膜病变(DR)患者58例为研究对象(PDR组),另选取同期因特发性黄斑裂孔行玻璃体切除术患者35例作为对照组。采用实时荧光定量(qRT-PCR)法检测玻璃体和膜组织中lncRNA PVT1、miR-26b、血管内皮生长因子(VEGF)mRNA表达水平,并使用全自动生化分析仪检测血清常规指标。结果PDR组血清空腹血糖(FBG)、糖化血红蛋白(HbA1c)水平明显高于对照组,高密度脂蛋白胆固醇(HDL-C)水平明显低于对照组(P<0.05)。PDR组患眼玻璃体和膜组织中lncRNA PVT1、VEGF mRNA表达水平明显高于对照组,miR-26b表达水平明显低于对照组(P<0.05)。PDR组患眼玻璃体和增生膜中lncRNA PVT1表达水平与miR-26b、患者血清HDL-C表达水平呈负相关(P<0.05),与患者血清FBG、HbA1c、VEGF mRNA呈正相关(P<0.05)。PDR患眼玻璃体和增生膜中miR-26b表达水平均与患者血清FBG、HbA1c、VEGF mRNA呈负相关,与患者血清HDL-C呈正相关(P<0.05)。结论PDR患眼玻璃体和增生膜中lncRNA PVT1水平显著升高、miR-26b水平显著降低,二者之间可能存在相互作用,共同影响PDR进展。Objective To investigate the expression levels and significance of long non-coding RNA plasmacytoma variant translocation 1 lncRNA PVT1 and micro RNA-26b(miR-26b)in the vitreous and proliferative membrane of eyes with proliferative diabetic retinopathy(PDR).Methods From March 2016 to March 2019,58 patients with uniocular diabetic retinopathy were selected as the PDR group,and 35 patients with idiopathic macular hole undergoing vitrectomy were selected as the control group.The expression levels of lncRNA PVT1,miR-26b and vascular endothelial growth factor(VEGF)mRNA in the vitreous and membranous tissues were detected by real-time fluorescence quantitative analysis(qRT-PCR),and the routine indexes of serum were detected by automatic biochemical analyzer.Results The levels of fasting blood glucose(FBG)and glycosylated hemoglobin(HbA1c)in the PDR group were significantly higher than those in the control group(P<0.05),and the level of high density lipoprotein cholesterol(HDL-C)was significantly lower than the control group(P<0.05).The expression levels of lncRNA PVT1 and VEGF mRNA in the vitreous and membrane tissue in the PDR group were significantly higher than the control group,while the expression level of miR-26b was significantly lower than the control group(P<0.05).The expression of lncRNA PVT1 in vitreous and proliferative membrane in the PDR group was negatively correlated with miR-26b and the expression level of serum HDL-C(P<0.05),while positively correlated with FBG,HbA1c and VEGF mRNA(P<0.05).The expression level of miR-26b in the vitreous and proliferative membrane in PDR patients was negatively correlated with FBG,HbA1c,VEGF mRNA,while positively correlated with serum HDL-C(P<0.05).Conclusions The level of lncRNA PVT1 in the vitreous and proliferative membrane in PDR patients is significantly increased,but the level of miR-26b is decreased significantly,possible correlations exist between these two factors,which will affect the progress of PDR.
