摘 要:目的探讨干扰素刺激基因-15(ISG15)、信号传导及转录激活因子-1(STAT1)在卵巢癌中的作用。方法从基因型组织表达(GTEx)数据库、肿瘤基因组图谱(TCGA)数据库下载并分析数据;利用基因集富集分析(GSEA)软件对通路富集进行分析;采用Perl语言整理数据,采用SPSS统计软件包进行统计分析,采用R语言进行数据分析、建立图片、京都基因与基因组百科全书(KEGG)-基因本体论(GO)富集分析。结果卵巢癌组织的ISG15表达水平明显高于正常卵巢组织(P<0.05);ISG15表达水平高的卵巢癌患者预后较好(P<0.05)。ISG15与STAT1呈正相关(r=0.629,P<0.05);卵巢癌组织的STAT1表达水平明显高于正常卵巢组织(P<0.05);STAT1高表达是卵巢癌的保护因子(P<0.05)。ISG15、STAT1均在RIG-Ⅰ样受体信号通路、溶酶体、TOLL样受体信号通路富集;同时,ISG15还富集于自噬调节,STAT1还富集于凋亡。结论ISG15及其密切相关基因STAT1在卵巢癌中可能发挥抑癌作用。Objective To investigate the role of interferon-stimulated genes-15(ISG15),signal transducers and activators of transcription-1(STAT1)in ovarian cancer.Methods Download and analyze the data from the Genotype-Tissue Expression(GTEx)database and The Cancer Genome Atlas(TCGA)database.The Gene Set Enrichment Assay(GSEA)software was used to analyze the pathway enrichment.Data was sorted out by Perl language,statistical analysis was conducted by SPSS software package,data analysis was conducted by R language,images were established,and enrichment analysis was conducted by Kyoto Encyclopedia of Kyoto Encyclopedia of Genes and Genomes(KEGG)-Gene Ontology(GO).Results The expression level of ISG15 in ovarian cancer tissues was significantly higher than that in normal ovarian tissues(P<0.05).Ovarian cancer patients with high ISG15 expression had a better prognosis(P<0.05).ISG15 was positively correlated with STAT1(r=0.629,P<0.05).The expression level of STAT1 in ovarian cancer tissues was significantly higher than that in normal ovarian tissues(P<0.05).High expression of STAT1 was a protective factor for ovarian cancer(P<0.05).ISG15,STAT1 in RIG-Ⅰreceptor signaling pathways,lysosome,toll-like receptors signaling pathway enrichment;ISG15 is also enriched in autophagy regulation and STAT1 is also enriched in apoptosis.Conclusion ISG15 and its closely related gene STAT1 may play an anticancer role in ovarian cancer.
