摘　　要：目的:研究去甲斑蝥素对人雄激素非依赖型前列腺癌PC-3细胞生长抑制与诱导凋亡的作用。方法:CCK-8技术检测去甲斑蝥素对PC-3细胞生长抑制率的影响,流式细胞仪检测去甲斑蝥素对PC-3细胞周期和凋亡率的影响。结果:去甲斑蝥素对PC-3细胞有生长抑制作用,随去甲斑蝥素浓度升高,抑制作用增强,呈现剂量-效应关系。流式细胞检测提示,去甲斑蝥素作用于PC-3细胞后,G 0/G 1期细胞明显减少,S、G 2期PC-3细胞增加,凋亡率升高(P<0.05)。结论:去甲斑蝥素能抑制人雄激素非依赖型前列腺癌PC-3细胞的增殖,促进PC-3细胞凋亡。Objective:To investigate the effect of norcantharidin on proliferation and apoptosis of human androgen independent prostate cancer cell line PC-3.Methods:CCK-8 method was used to detect the growth inhibition rate of PC-3 cells,and flow cytometry was used to detect the cell cycle and the apoptosis rate of PC-3 cells.Results:Norcantharidin could inhibit the growth of PC-3 cells.With the increase of norcantharidin concentration,the effect increased in a dose-response relationship.Flow cytometry showed that after norcantharidin treated PC-3 cells,G 0/G 1 phase cells decreased,S and G 2 phase cells increased,apoptosis rate increased(P<0.05).Conclusion:Norcantharidin can significantly inhibit the proliferation of human androgen independent prostate cancer cell line PC-3 and induce it apoptosis.
