摘 要:2肾上腺素能受体(Beta-2 adrenergic receptor)是一种G蛋白偶联受体(G-protein coupled receptors),与生物体的生长、内分泌和代谢等生理活动有着密切的联系。本研究从缢蛏(Sinonovacula constricta)转录组文库中筛选得到缢蛏β2肾上腺素能受体的片段序列。通过PCR和RACE技术获得β2肾上腺素能受体的cDNA全长序列,命名为ScADRB2,其长度为1 497 bp,包括148 bp的5’UTR、191 bp的3’UTR以及1 158 bp的开放阅读框,编码385个氨基酸。通过实时荧光定量PCR检测ScADRB2 m RNA在血淋巴、外套膜、斧足、鳃、肝胰腺、性腺和水管的表达量。结果表明,该基因在外套膜的表达量最高,其次为斧足,在水管和性腺中的表达量较低,而在血淋巴、肝胰腺和鳃中的表达量最低。设计组织损伤实验,损伤缢蛏的进水管,在处理后第8小时、第24小时、第36小时、第48小时、第60小时和第72小时分别取样。通过荧光定量PCR分析,结果表明,其在8 h和24 h时表达量较高,在36 h时表达量显著下调,在48 h时表达量有所上升,在60 h、72 h又有所下调。研究结果表明,缢蛏的β2肾上腺素能受体可能参与了损伤修复的过程。Beta-2 adrenergic receptor is a G protein coupled receptor, which is closely related to biological activities such as growth, endocrine and metabolism. The sequence of beta-2 adrenergic receptor was screened from the transcriptional library of the razor clam Sinonovacula constricta, and the full-length sequence of beta-2 adrenergic receptor was obtained by PCR and RACE technology, named as ScADRB2. The cDNA length of ScADRB2 was 1 497 bp, including 148 bp 5’ untranslated region, 191 bp 3’ untranslated region, and 1 158 bp open reading frame, encoding 385 amino acids. Primers were designed in ORF to detect the expression level in hemolymph,liver, waterpipe, foot, mantle, gill, and gonad, by quantitative real-time PCR, respectively. The results showed that the expression of ScADRB2 was the highest in the mantle, relatively high in the foot, lower in the siphon and gonad, but the lowest in the hemolymph, liver and gills. Tissue injury experiment was designed and the samples were collected at 8 h, 24 h, 48 h, 60 h and 72 h after injuring siphon. The results showed that the expression level of ScADRB2 was higher at 8 h and 24 h, decreased significantly at 36 h, increased at 48 h, and decreased at 60 h or 72 h. The results showed that β-2 adrenergic receptor might be involved in the process of damage and repair of S. constricta.
