摘 要:目的研究苦豆碱(ALO)对血小板源生长因子BB(PDGF-BB)刺激建立的人肺动脉平滑肌细胞(HPASMCs)增殖模型的抗氧化作用。方法通过采用20 ng/mL PDGF-BB刺激构建人肺动脉平滑肌细胞增殖模型,给予苦豆碱0.125 mM、0.25 mM、0.5 mM干预。通过采用比色法来检测HPASMCs中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)、丙二醛(MDA)的活性以及总抗氧化能力(T-AOC)。结果与正常对照组相比,PDGF-BB诱导的HPASMCs中,MDA水平显著升高,SOD、CAT、GSH-PX以及总抗氧能力的活性显著降低。予苦豆碱干预后,与PDGF-BB诱导的HPASMCs比较,苦豆碱干预组HPASMCs中MDA含量明显降低,细胞中SOD、CAT、GSH-Px以及总的抗氧能力活性显著升高。结论苦豆碱对PDGF-BB刺激构建人肺动脉平滑肌细胞增殖模型具有抗氧化作用。Objective To investigate the antioxidant effects of Aloperine(ALO)on the proliferation of human pulmonary artery smooth muscle cells(HPASMCs)induced by platelet derived growth factor-BB(PDGF-BB).Methods The HPASMCs model was established by 20 ng/mL PDGF-BB and the cells were intervened with ALO of 0.125 mM,0.25 mM and 0.5 mM.The activity of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-PX),malondialdehyde(MDA)and the total antioxidant capacity(T-AOC)were measured in HPASMCs by colorimetry method.Results Compared with the control group,the MDA level was significantly increased in the PDGF-BB-induced HPASMCs model group,and the activities of SOD,CAT,GSH-PX and total antioxidant capacity were significantly decreased.After intervention with ALO,compared with the model group,the content of MDA in smooth muscle cells of ALO group was significantly decreased,and the activity of SOD,CAT,GSH-PX and T-AOC were significantly increased.Conclusion ALO has the antioxidant effect against the proliferation model of HPASMCs induced by PDGF-BB.
