摘 要:目的探讨哺乳动物雷帕霉素靶蛋白(mTOR)信号通路相关分子在肺鳞癌组织中的表达情况及其与临床病理特征、生存期的关系,寻找治疗的新靶点。方法收集2010年6月至2017年12月在舟山医院经手术病理证实的肺鳞癌组织标本93例,采用免疫组织化学方法检测mTOR信号通路相关分子的表达情况,分析各分子与临床病理特征及生存期关系。结果10号染色体缺失的磷酸酶及张力蛋白同源物(PTEN)低表达患者中,肿瘤直径>3 cm患者所占比例为89.1%(49/55),与高表达患者比较差异有统计学意义(P<0.05)。真核细胞翻译起始因子4E结合蛋白1(4EBP1)高表达患者中,淋巴结发生转移与高于Ⅱ期患者所占比例分别为47.5%(29/61)与80.3%(49/61),与低表达患者比较差异均有统计学意义(均P<0.05)。p-mTOR高表达患者中,患者整体生存期较短(35.0个月),与低表达患者比较差异有统计学意义(P<0.05)。结论mTOR信号通路相关分子有可能成为肺鳞癌诊断指标或作为临床治疗的靶点。Objective To investigate the expression of mammalian target of rapamycin(mTOR)signaling pathway-related proteins in lung squamous cell carcinoma and its relation to clinicopathological features and survival of patients.Methods A total of 93 patients with lung squamous cell carcinoma confirmed by surgery and pathology in Zhoushan hospital from June 2010 to December 2017 were enrolled in the study.The expression of proteins related to mTOR signaling pathway was detected by immunohistochemical method,and the relationship between protein expression and clinical characteristics and survival of patients was analyzed.Results In patients with lower PTEN expression,the proportion of tumor diameter>3 cm was higher(89.1%,49/55)than those with high PTEN expression(P<0.05).Patients with high expression of 4EBP1 had higher rates of lymph node metastasis(47.5%,29/61)and phaseⅡdisease(80.3%,49/61)(P<0.05).Patients with higher p-mTOR expression had shorter overall survival time(35.0 months)(P<0.05).Conclusion The key signaling molecules in the mTOR signaling pathway might be used as indicators for the diagnosis of lung squamous cell carcinoma or as a clinical therapeutic target.
