益肾固本膏方对肾阳虚哮喘模型大鼠ACTH、CORT的影响

摘　　要：目的观察益肾固本膏方对肾阳虚哮喘模型大鼠血浆促肾上腺皮质激素(ACTH)、皮质酮(CORT)水平的影响,进一步探讨益肾固本膏方改善肾阳虚哮喘患者肾阳虚状态的具体机制。方法将SD大鼠分为空白对照组、肾阳虚哮喘模型组、益肾固本膏方低、中、高剂量组。采用氢化可的松琥珀酸钠、Al(OH)3及吸入卵蛋白制备肾阳虚哮喘大鼠模型。观察动物一般状态及其体质量变化,采用Elisa法检测大鼠血中ACTH、CORT的含量。结果肾阳虚哮喘模型组大鼠体质量明显低于空白对照组(P<0.05)。益肾固本膏方各剂量组ACTH及CORT含量均比肾阳虚哮喘模型组显著升高(P<0.01)。结论肾阳虚哮喘模型大鼠HPA轴功能低下,益肾固本膏方能拮抗肾阳虚哮喘HPA功能之低下,有效改善哮喘患者“肾虚”的病理状态,增加内源性皮质酮含量,减少其哮喘急性发作的频率和严重程度。Objective To observe the effect of Yishen Guben ointment on plasma adrenocorticotropic hormone(ACTH)and corticosterone(CORT)levels in rats with kidney-yang deficiency asthma,and further explore the mechanism of Yishen Guben ointment on improving kidney-yang deficiency of patients with asthma.Methods SD rats were divided into normal control group,kidney-yang deficiency asthma model group,Yishen Guben cream prescription low,medium and high dose group,prepared with hydrocortisone sodium succinate,Al(OH)3 and inhaled egg albumin.Rats with kidney-yang deficiency asthma were observed.The general state of the animals and their weight were observed.The content of ACTH and CORT in the blood of rats was detected by Elisa method.Results The weight of rats in the kidney-yang deficiency asthma model group was significantly lower than that in the blank control group(P<0.05).The levels of ACTH and CORT in Yishen Guben Ointment group were significantly higher than those in the kidney-yang deficiency asthma model group(P<0.01).Conclusion In the kidney-yang deficiency asthma model,the HPA axis function is low.Yishen Guben ointment can antagonize the low function of HPA axis in kidney yang deficiency asthma,effectively improve the pathological state of“kidney deficiency”in asthma patients,increase endogenous corticosterone content,and reduce its frequency and severity of acute asthma attacks.
