摘  要:目的探讨19号染色体开放阅读框5(C19ORF5)蛋白在结直肠癌组织中的表达情况,分析C19ORF5蛋白调节自噬活性与结直肠癌恶性程度的关系。方法收集141例结直肠癌患者的结直肠癌组织和癌旁正常组织,分别采用免疫组织化学染色方法和实时定量聚合酶链反应(PCR)检测两种组织中C19ORF5蛋白和mRNA表达情况,在电镜下观察组织的超微结构,分析C19ORF5蛋白对自噬活性的调节作用,比较不同临床特征结直肠癌患者结直肠癌组织中C19ORF5蛋白的表达情况。结果结直肠癌组织中C19ORF5蛋白的染色强度明显高于癌旁正常组织,Ⅰ/Ⅱ期结直肠癌组织中C19ORF5蛋白的染色强度明显高于Ⅲ/Ⅳ期结直肠癌组织,Ⅰ/Ⅱ期结直肠癌组织的自噬活性较高,Ⅲ/Ⅳ期结直肠癌组织的自噬活性较低。结直肠癌组织中C19ORF5 mRNA的表达水平明显高于癌旁正常组织,差异有统计学意义(P﹤0.01)。TNM分期为Ⅰ~Ⅱ期、癌胚抗原(CEA)升高、无肝转移结直肠癌患者结直肠癌组织中C19ORF5蛋白的高表达率分别高于TNM分期为Ⅲ~Ⅳ期、CEA正常、有肝转移的患者,差异均有统计学意义(P﹤0.05)。结论在结直肠癌组织中,高表达的C19ORF5蛋白可增加肿瘤细胞的自噬活性,增加基因的稳定性,降低肿瘤的恶性程度。C19ORF5蛋白调节自噬活性与结直肠癌的恶性程度呈负相关,C19ORF5可能是一种重要的抑癌基因。Objective To investigate the expression level of chromosome 19 open reading frame 5(C19ORF5) in colorectal cancer and analyze its regulatory function on autophagic activity and its relationship with the malignant degree of colorectal cancer. Method A total of 141 colorectal cancer tissues and 141 adjacent normal tissues were collected.The immunohistochemistry assay and quantitative real-time polymerase chain reaction(PCR) were employed to detect the expression of C19ORF5 protein and mRNA respectively in colorectal cancer tissues and adjacent normal tissues and their ultrastructure was observed under the electron microscope. The regulatory function of C19ORF5 on autophagic activity were analyzed, and the expression level of C19ORF5 protein was compared between patients with different clinical characteristics. Result The staining intensity of C19ORF5 protein in colorectal cancer tissues was significantly higher than that in adjacent normal tissues, and the staining intensity of C19ORF5 protein was higher in stage Ⅰ/Ⅱ colorectal cancer tissues than that in stage Ⅲ/Ⅳ colorectal cancer tissues. Samples from patients with stage Ⅰ/Ⅱ colorectal cancer showed higher autophagic activity than those from patients with stage Ⅲ/Ⅳ colorectal cancer. The expression of C19ORF5 mRNA in colorectal cancer tissues was higher than that in adjacent normal tissues, with statistically significant difference(P<0.01). The high expression rate of C19ORF5 protein was significantly higher in patients with Ⅰ-Ⅱ stage, elevated carcinoembryonic antigen(CEA) and no liver metastasis than patients with Ⅲ-Ⅳ stage, normal CEA level and liver metastasis, the difference was statistically significant(P<0.05). Conclusion High expression of C19ORF5 protein in colorectal cancer tissues could strengthen the autophagic activity of cancer cells, increase the stability of genes and reduce the malignant degree of cancer.
