摘 要:本实验旨在研究牦牛不同肌肉组织的肉品质和脂肪代谢相关基因表达水平,并分析肌内脂肪(IMF)含量与脂肪代谢相关基因表达的相关性。选用4岁健康麦洼牦牛10头,屠宰后采集小黄瓜条、辣椒条和外脊测定IMF含量、剪切力和脂肪代谢相关基因表达水平,并分析基因表达水平与IMF含量的相关性。结果表明:外脊的IMF含量显著高于小黄瓜条和辣椒条,小黄瓜条的剪切力显著高于辣椒条,且二者均显著高于外脊;外脊的硬脂酰辅酶A去饱和酶1(SCD1)、固醇调节元件结合蛋白-1(SREBP-1)、脂肪酸合成酶(FAS)、过氧化物酶增殖激活受体γ(PPARγ)基因表达显著高于小黄瓜条和辣椒条,辣椒条的SCD1、SREBP-1、FAS、PPARγ基因表达显著高于小黄瓜条,外脊的激素敏感脂肪酶(HSL)和肉碱转移酶1(CPT-1)基因表达显著低于小黄瓜条,外脊的CPT-1基因表达显著低于辣椒条,辣椒条的HSL基因表达显著低于小黄瓜条;小黄瓜条的SCD1基因表达与IMF含量呈极显著正相关,辣椒条和外脊的SCD1基因表达与IMF含量呈显著正相关,小黄瓜条、辣椒条和外脊的HSL、CPT-1基因表达与IMF含量呈显著负相关。结果提示,与小黄瓜条和辣椒条相比,外脊通过促进脂肪合成相关基因(SCD1、SREBP-1、FAS和PPARγ)的表达以及抑制脂肪分解相关基因(HSL和CPT-1)的表达,促进IMF沉积,提高牛肉嫩度,改善牦牛肉品质;并且SCD1、HSL和CPT-1基因可作为调控牦牛肌肉IMF形成的候选基因,为进一步揭示IMF沉积机制及优化牦牛肉品质奠定基础。The purpose of this study was to study the meat quality and the expression of genes related to fat metabolism in different muscle tissues of yaks, and to analyze the relationship between the content of fat(IMF) content and the expression of genes related to fat metabolism. Ten healthy 4-year-old Maiwa yaks were selected. Eye round, chuck tender and striploin were collected after slaughter, and IMF content and shear force were measured. Meanwhile, gene expression levels related to fat metabolism were detected, and the correlation between gene expression levels and IMF content was analyzed. The results showed that IMF content in striploin was significantly higher than that in eye round and chuck tender(P<0.05). The shear force of eye round and chuck tender was significantly higher than that of the striploin(P<0.05). The shear force of eye round was significantly higher than that of chuck tender(P<0.05). The expression of stearoyl-CoA desaturase 1(SCD1), sterol regulatory element binding protein 1(SREBP-1), fatty acid synthase(FAS) and peroxisome proliferator-activated receptor γ(PPARγ) genes in the striploin was significantly higher than that in eye round and chuck tender(P<0.05). The expression of SCD1, SREBP, FAS and PPARγ genes in chuck tender was significantly higher than that in eye round(P<0.05). The expression of hormone-sensitive lipase(HSL) and carnitine palmitoyltransferase-1(CPT-1) genes in the striploin was significantly lower than that in the eye round(P<0.05). CPT-1 gene expression in the striploin was significantly lower than that in chuck tender(P<0.05). The HSL gene expression in chuck tender was significantly lower than that in eye round(P<0.05). There was a significant positive correlation between SCD1 gene expression and IMF content in eye round(P<0.01). There was a significant positive correlation between SCD1 gene expression in chuck tender and striploin and IMF content(P<0.05).
