摘  要:本试验旨在探究魏斯氏乳杆菌(L. weissella)、唾液乳杆菌(L. salivarius)和约氏乳杆菌(L.johnsonii)联合壳聚糖季铵盐(CQN)对奶牛子宫内膜炎的影响。通过乳酸脱氢酶(LDH)释放检测法筛选致病性大肠杆菌(EPEC)最佳处理浓度和时间,建立奶牛子宫内膜上皮细胞炎症模型。诱导细胞炎症后,单独添加0.01 mg/mL CQN、108CFU/mL乳杆菌处理以及乳杆菌联合CQN处理,ELISA法和RT-qPCR法检测细胞因子IL-6、IL-8和IL-10的变化。结果表明:单独添加3株乳杆菌处理与单独添加CQN处理均降低了IL-6的蛋白和mRNA表达(P<0.05),但对IL-8的表达无显著影响;相比单独添加处理,约氏乳杆菌与CQN混合处理降低了IL-6和IL-8的表达(P<0.05),并恢复了IL-6/IL-10和IL8/IL-10的平衡;与CQN混合处理后,约氏乳杆菌对奶牛子宫内膜上皮细胞的黏附率提升(P<0.001)。结果提示,约氏乳杆菌联合CQN具有良好的抗炎潜力,可用于奶牛子宫内膜炎的治疗。The purpose of this study is to investigate the effects of three strains of L. weissella, L. salivarius, and L. johnsonii combined with chitosan quaternary ammonium salt(CQN) on cow endometritis. The concentration and time of cells treated by E. coli were screened by lactate dehydrogenase(LDH) release detection method, relying to which we established a cell inflammation model. After cell inflammation induced, the cells were treated by adding 0.01 mg/mL CQN alone, 108 CFU/mL Lactobacillus alone, and Lactobacillus combined with CQN. After that, expression of cytokines IL-6, IL-8 and IL-10 were detected by ELISA and RT-qPCR. The results showed that the addition of three strains of Lactobacillus alone and the addition of CQN alone significantly reduced the protein and mRNA expression of IL-6(P<0.05), but showed no significant effect on the expression of IL-8. However, compared with the single addition treatment, the mixed treatment of L. johnsonii and CQN significantly reduced the expression of IL-6 and IL-8(P<0.05) and restored the balance of IL-6/IL-10 and IL8/IL-10. At the same time, after mixed treatment with CQN, the adhesion rate of L. johnsonii to cow endometrial epithelial cells was significantly improved(P<0.001). The results suggested that alliance of L. johnsonii and CQN showed good anti-inflammatory potential, and it could be used in treat for cow endometritis.
