摘　　要：目的:探讨miR-425-5p通过下调PTEN表达对乳腺癌细胞增殖、迁移及侵袭的影响及其潜在的作用机制。方法:运用qPCR检测乳腺癌细胞MCF-7、MDA-MB-231和正常乳腺细胞MCF-10A中miR-425-5p和PTEN的mRNA表达水平,Western blot检测PTEN蛋白表达水平。分别建立过表达miR-425-5p、抑制miR-425-5p表达或过表达PTEN的MCF-7和MDA-MB-231细胞株,采用MTT法检测细胞增殖活力,Transwell小室检测细胞的迁移及侵袭能力;双荧光素酶报告基因分析法和Western blotting验证miR-425-5p和PTEN的靶向调控关系。结果:与正常乳腺细胞MCF-10A相比,乳腺癌细胞MCF-7和MDA-MB-231中miR-425-5p的表达显著升高(P<0.05),PTEN的表达显著降低(P<0.05)。过表达miR-425-5p促进MCF-7和MDAMB-231细胞的增殖、迁移及侵袭(P<0.05)。抑制miR-425-5p或过表达PTEN均可抑制MCF-7和MDA-MB-231细胞的增殖、迁移及侵袭(P<0.05)。PTEN是miR-425-5p的靶基因,miR-425-5p可负性调控PTEN的表达。结论:miR-425-5p可通过下调PTEN蛋白的表达,从而促进乳腺癌细胞的增殖、迁移及侵袭能力。Objective:To investigate the effect and potential mechanismof miR-425-5p on the proliferation,migration and invasion of breast cancer cells by down-regulating the expression of PTEN.Methods:The mRNA expression of miR-425-5p and PTEN in breast cancer cells MCF-7,MDA-MB-231 and normal breast cell MCF-10A was detected by qPCR.The expression level of PTEN protein was detected by Western blot.MCF-7 and MDA-MB-231 cell lines of overexpression miR-425-5p,inhibition of the expression of miR-425-5p or overexpression of PTEN were established.The cell proliferation was detected by MTT assay.The migration and invasion ability was detected by Transwell chamber.The targeted regulation relationship between miR-425-5p and PTEN was verified by double luciferase reporter gene assay and Western blot.Results:The expression of miR-425-5p in breast cancer cells MCF-7 and MDA-MB-231 was significantly higher than that in normal breast cells MCF-10A(P<0.05).The expression of PTEN was significantly decreased(P<0.05).Overexpression of miR-425-5p promoted the proliferation,migration and invasion of MCF-7 and MDA-MB-231 cells(P<0.05).Inhibition of miR-425-5p or overexpression of PTEN could inhibit the proliferation,migration and invasion of MCF-7 and MDAMB-231 cells(P<0.05).PTEN was the target gene of miR-425-5p,and miR-425-5p negatively regulated the expression of PTEN.Conclusion:MiR-425-5p can promote the proliferation,migration and invasion of breast cancer cells by down-regulating the expression of PTEN protein.
