摘 要:目的:分析氟西汀对弱视成年大鼠视皮层17区N-甲基-D-天冬氨酸受体亚基(NMDA-NR)的影响,探讨氟西汀对弱视视皮层突触可塑性的作用。方法:选取45只2周龄Wistar大鼠,其中15只大鼠不作任何特殊处理,仅给予标准饲养,作为对照组;另外30只在整个关键期内对右侧眼睑进行缝合,制作单眼形觉剥夺(MD)弱视模型。45日龄时将30只模型大鼠随机分为模型组与氟西汀组(每日喂养0.2 mg/mL氟西汀水),每组15只。上述3组大鼠均在60日龄(T1)、75日龄(T2)、105日龄(T3)时取5只处死,并观察不同时刻各组大鼠的视皮层17区NMDA-NR2A、NMDA-NR2B表达、C/I值(左眼/右眼幅值)、图形视觉诱发电位(PVEP)振幅值。结果:T1、T2、T3时刻各组的视皮层17区NMDA-NR2A、NMDA-NR2B表达量比较,差异有统计学意义(P<0.05);同一时刻对照组和氟西汀组的视皮层17区NMDA-NR2A、NMDA-NR2B表达量均高于模型组,且对照组高于氟西汀组(P<0.05);T1时刻对照组的C/I值低于模型组和氟西汀组,T2、T3时刻氟西汀组的C/I值均低于对照组和模型组(P<0.05);T1时刻各组的PVEP振幅值比较,差异无统计学意义(P>0.05);T2、T3时刻模型组的PVEP振幅值均低于对照组和氟西汀组,且氟西汀组低于对照组(P<0.05)。结论:关键期后给予弱视成年大鼠氟西汀治疗,可能通过改变视皮层中NMDANR2A、NMDA-NR2B的表达,激活视皮层可塑性,从而改善弱视。Objective:To analyze the effect of fluoxetine on N-methyl-D-aspartate receptor subunit(NMDANR)in visual cortex of adult rats with amblyopia,and to explore the effect of fluoxetine on synaptic plasticity of amblyopic cortex.Methods:A total of 452-week-old Wistar rats were selected,of which 15 rats were given standard feeding without any special treatment and were taken as control group,and the other 30 rats were sutured on the right eyelid in the whole critical period to make monocular deprivation(MD)amblyopia model.At the age of 45 days,30 model rats were randomly divided into model group and fluoxetine group(fed with 0.2mg/mL fluoxetine water daily),with 15 rats in each group.Rats in the above three groups were sacrificed at 60 days old(T1),75 days old(T2)and 105 days old(T3).The expression of NMDA-NR2A and NMDA-NR2B,C/I value(left eye/right eye amplitude)and amplitude of pattern visual evoked potential(PVEP)were observed in each group at different time.Results:There were significant differences in the expression of NMDA-NR2A and NMDA-NR2B in 17 area of visual cortex among different groups at T1,T2 and T3(P<0.05).At the same time,the expression of NMDA-NR2A and NMDA-NR2B in 17 area of visual cortex in control group and fluoxetine group was higher than that in model group,and that of the control group was higher than that of the fluoxetine group(P<0.05).The C/I value of the control group at T1 was lower than that of the model group and fluoxetine group,and the C/I value of the fluoxetine group at T2 and T3 was lower than that of the model group and control group(P<0.05).There was no significant difference in PVEP amplitude among each group at T1(P>0.05).PVEP amplitude at T2 and T3 in the model group was lower than that in the control group and fluoxetine group,and that in the fluoxetine group was lower than that in the control group(P<0.05).Conclusion:Fluoxetine treatment after the critical period in adult rats with amblyopia may improve amblyopia by changing the expression of NMDA-NR2A and NMDA-NR2B in the visua
