摘  要:目的探究雷帕霉素(RAPA)对人脐静脉血管内皮细胞(HUVECs)放疗的防护作用。方法体外培养第3代人脐静脉血管内皮细胞,接受8Gy单次放疗,观察不同浓度(0、 0.1、1、 20 nmol/L)雷帕霉素对人脐静脉血管内皮细胞放疗后的增殖,凋亡和周期的影响,并检测HUVEC mTOR,DNA损伤标志物(γH2AX),活性氧自由基(ROS)清除剂锰过氧化物歧化酶(MnSOD)等表达情况。结果 HUVECs加载雷帕霉素在细胞放疗后增殖能力显著增强,凋亡比例明显降低。同时,检测发现雷帕霉素抑制了细胞放疗后mTOR的激活,通过高表达细胞内MnSOD,使γH2AX表达明显降低。结论雷帕霉素通过增加HUVEC MnSOD表达水平,降低γH2AX的表达,有效地减轻HUVECs细胞放疗后的DNA损伤反应,防止细胞进入衰老状态,对人血管内皮细胞放疗具有防护作用。Objective To investigate protection of rapamycin on human umbilical vein endothelial cells under irradiation-induced injury.Methods The third generation of human umbilical vein endothelial cells were cultured in vitro,and exposed to one single dose(8 Gy)irradiation.The effect of different concentration of rapamycin on cell proliferation,apoptosis and cell cycle after irradiation was evaluated.The expressions of mTOR pathway,MnSOD andγH2AX were examined.Results Rapamycin protected the proliferation and decreased cell apoptosis of HUVECs after irradiation.Meanwhile,rapamycin completely blocked radiation-induced mTOR activation and downregulated the expression ofγH2AX by increased expression of MnSOD.Conclusion Rapamycin reduced accumulation of DNA damage response markerγH2AX by increased expression of MnSOD and protected HUVECs by decreasing ROS-induced DNA damage response after irradiation.
