摘  要:采用网络药理学研究方法,探讨头痛宁治疗蛛网膜下腔出血(subarachnoid hemorrhage,SAH)的分子机制。利用中药相关数据库搜索出头痛宁的有效化学成分,这些成分按照口服生物利用度(oral bioavailability,OB)、类药性(Drug-likeness,DL)及血脑屏障(blood-brain barrier,BBB)透过性差异分类,并找到对应靶点。应用Cytoscope软件"merge"功能将药物靶点与SAH的疾病靶点匹配,得到治疗SAH的关键靶点,经Metascape平台进行富集分析。结果表明根据OB≥30%、DL≥0.18及血脑屏障(blood brain barrier,BBB)≥0.3或BBB<0.3筛选标准,得到头痛宁容易透过BBB的7种有效成分及不易透过BBB的14种有效成分,它们分别对应着57、120个靶点。将这些靶点与SAH的273个靶点共同分析后得到易透过BBB的10个关键节点及不易透过BBB的28个关键节点,Metascape分析结果显示这些关键节点主要在炎症、神经元修复、神经递质调节、周围神经重塑、脂质及激素代谢、血管舒缩调节方面富集。揭示了头痛宁治疗蛛网膜下腔出血的作用机制,体现了中药复方多靶点的治疗优势,为进一步药理学研究提供了实验依据。The purpose of this study was to explore the therapeutic mechanism of Toutongning in the treatment of subarachnoid hemorrhage(SAH)based on the network pharmacology approaches.This studylisted the chemical compounds of Toutongning,which were classified according to the difference of oral bioavailability(OB),drug-like(DL)and blood brain barrier(BBB)permeability,and the corresponding targets were found out.Then,those targets werematched with the disease targets of SAH and we found outthe key targets for the treatment of SAH.Finally,the enrichment analysis was performed by metascape platform.According to OB,DL and BBB≥0.3 or BBB<0.3,our study found out 7 potential active compounds which were easily across the BBB and 14 compounds which were not.And those compounds corresponded to 57 and 120 targets respectively.These targets were analyzed together with 273 targets of SAH to obtain 10 key nodes which were easy to across BBB and 28 key nodes which were not.Metascape analysis showed that these nodes were mainly enriched in inflammation,neuron repair,neurotransmitter regulation,peripheral nerve remodeling,lipid and hormone metabolismand vasomotor regulation.This studyrevealed the potential mechanism of Toutongning in the treatment of SAH and built a systematical network pharmacology approach,indicating that the traditional Chinese medicine hasadvantages of multiple targets therapy of SAH,and our results provided a basis for the future study of Toutongning.
