摘 要:目的探讨葫芦素Ⅰ对咪喹莫特诱导的银屑病小鼠模型的影响及相关作用机制。方法30只BALB/c雌性小鼠随机分为3组,空白组、模型组、实验组。用PASI评分动态观察小鼠皮损变化,光镜下观察小鼠皮损病理改变并测量表皮厚度。用酶联免疫吸附法检测各组小鼠血清中IL-17、IL-22、IFN-γ的含量变化,免疫组织化学检测小鼠皮损中STAT3、p-STAT3的含量表达,实时荧光定量PCR法检测小鼠皮损中STAT3和K17的mRNA表达。结果空白组小鼠皮肤未见明显改变。与模型组相比,实验组小鼠红斑、鳞屑严重程度减轻,PASI评分和血清炎症细胞因子水平明显下降(P<0.05)。免疫组织化学显示,皮损中STAT3、p-STAT3表达较模型组低,STAT3和K17的mRNA表达含量也下降,差异均有统计学意义(P均<0.05)。结论葫芦素Ⅰ可抑制STAT3磷酸化,一定程度上缓解小鼠银屑病皮损,减轻炎症反应,并在mRNA水平下调STAT3和K17的表达。Objective To investigate the effect of cucurbitacin Ⅰ on mouse model of psoriasis induced by imiquimod and its related mechanism. Methods Thirty BALB/c female mice were randomly divided into 3 groups:blank group,model group and experimental group.Psoriasis area severity index(PASI) was scored to measure the changes of skin lesions in mice.Optical microscope to observed pathological changes of skin lesions and to measure the skin thickness of epidermis.The changes of IL-17,IL-22 and IFN-γ in mouse serum in each group were detected by ELISA.Immunohistochemical study to evaluate changes of STAT3 and p-STAT3 levels in skin lesions.Real-time fluorescence-based quantitative PCR(qRT-PCR) was performed to determine the mRNA expression of STAT3 and K17 in mouse skin lesions.Results The skin of mice in the blank group did not change significantly.Compared with the model group,the severity of erythema and scales in the experimental group were reduced,PASI score and inflammatory cytokines in serum were significantly decreased(P<0.05).Immunohistochemistry showed that the expressions of STAT3 and p-STAT3 in the lesions lower than the model group,and the mRNA expressions of STAT3 and K17 were also decreased,the difference was statistically significant(all P<0.05).Conclusion Cucurbitin Ⅰ can inhibited the phosphorylation of STAT3,relieve the psoriasis-like lesions in mice to a certain extent,reduced the inflammatory response,and down-regulate the mRNA expression of STAT3 and K17.
