摘  要:为优化超声细胞破碎机辅助低共熔溶剂提取桑叶1-脱氧野尻霉素(DNJ)提取工艺,以DNJ提取量为指标,筛选低共熔溶剂的种类,通过单因素试验评价氯化胆碱和甘油的摩尔比、含水量、超声时间对DNJ提取量的影响,利用响应面法对影响DNJ提取的3个因素进行优化。结果表明,氯化胆碱甘油和水组成低共熔溶剂提取效果最好,最佳提取工艺条件为:氯化胆碱甘油摩尔比1︰3.4、含水量40%、超声时间32 min,重复3次,DNJ提取量为1.445±0.0018mg/g与模型预测值1.447 mg/g接近,模型能较准确预测桑叶DNJ提取量。In order to optimize the extraction process of mulberry leaf 1-deoxynojirimycin(DNJ)by ultrasonic cell crusher,the extraction amount of DNJ was used as the index to screen the types of deep eutectic solvents.The effects of molar ratio of choline chloride and glycerol,water content and ultrasonic time on the extraction amount of DNJ were investigated by single factor test.The response surface method was used to optimize the three factors affecting the extraction of DNJ.The results showed that the optimum extraction condition was choline chloride-glycerol ratio of 1︰3.4,water content of 40%,ultrasonic time of 32 min,and three parallel experiments.The average extraction amount of mulberry leaf DNJ was 1.445±0.0018 mg/g was close to the model prediction value of 1.447 mg/g.The model could accurately predict the extraction amount of mulberry DNJ.
