摘 要:目的探讨艾司西酞普兰(ESC)体外对大鼠海马神经干细胞(NSCs)存活和增殖的影响。方法取4只新生SD大鼠两侧海马组织,将其制成单细胞悬液,加入无血清的NSCs培养基培养,将稳定传代后的海马NSCs分为对照组和ESC组,对照组仍采用无血清的NSCs培养基继续培养;ESC组中加入不同浓度(5、10、20、40μmol/L)ESC进行干预。通过MTS检测不同浓度ESC对NSCs存活率的影响,筛选最有益于NSCs存活的ESC浓度,将其用于后续试验研究。采用细胞生长曲线和台盼蓝染色观察细胞增殖和存活情况,免疫荧光染色法和RT-PCR技术检测NSCs中的巢蛋白(Nestin)表达。结果稳定传代后的神经球呈规则立体球体,神经球内大多数细胞呈Nestin染色阳性。筛选发现浓度20μmol/L ESC最有益于NSCs的存活。MTS检测显示,对照组及20μmol/L ESC组1~5 d的细胞存活率比较无统计学差异。细胞生长曲线显示,对照组及20μmol/L ESC组NSCs的生长呈持续增殖趋势,20μmol/L ESC组的细胞数目较对照组多(P<0.05),增殖速率较对照组高(P<0.05)。免疫荧光染色结果显示,20μmol/L ESC组Nestin阳性神经球个数较对照组多(P<0.05)。RT-PCR结果显示,20μmol/L ESC组Nestin mRNA表达较对照组高(P<0.05)。结论ESC体外对大鼠海马NSCs具有促进存活和增殖的作用。Objective To study the effects of escitalopram(ESC)on the survival and proliferation of neural stem cells(NSCs)derived from hippocampus in newborn SD rats.Methods The hippocampus of 4 newborn SD rats were prepared into single-cell suspension and cultured in serum-free NSCs medium.The NSCs were divided into the control group and ESC group.The NSCs in the control group were still cultured in serum-free NSCs medium;the NSCs in the ESC group were added with different concentrations(5,10,20,40μmol/L)of ESCs for culture.The effects of different concentrations of ESCs on the survival rate of hippocampal NSCs were detected by MTS.The concentration of ESC(20μmol/L)was screened out to be the most favorable for the survival of NSCs and was used in the subsequent studies.Cell growth curve and Trypan blue staining were used to observe cell proliferation and survival.Immunofluorescence and RT-PCR were used to observe the expression changes of Nestin in NSCs.Results After stable passage,the nerve balls presented regular round shape,most of the cells in the nerve balls presented positive Nestin staining.MTS showed that there was no significant difference in cell survival rates on day 1-5 between the control group and 20μmol/L ESC group.The cell growth curve showed that the NSCs in the control group and the 20μmol/L ESC group showed a trend of continuous proliferation,and the number of cells in the 20μmol/L ESC group was higher than that in the control group(P<0.05),as well as the proliferation rate(P<0.05).The number of Nestin positive nerve balls increased in the ESC group(20μmol/L)as compared with that of the control group(P<0.05).RT-PCR showed that the expression of Nestin in the 20μmol/L ESC group was significantly higher than that in the control group(P<0.05).Conclusion ESC can promote the survival and proliferation of hippocampal NSCs.
