摘 要:目的观察大鼠坐骨神经慢性卡压损伤对背根神经节纤维化的影响,并探讨背根神经节纤维化的相关机制。方法按照Mackinnon设计的方法将30只SD大鼠建立大鼠坐骨神经慢性卡压模型,将右侧坐骨神经手术侧设为卡压侧,左侧假手术侧设为对照侧。造模3周和8周时,分别切取卡压侧、对照侧坐骨神经支配的腓肠肌,行Masson染色,测算腓肠肌横截面积(CSA)以及胶原容积分数(CVF)。造模3周时,分别取大鼠坐骨神经卡压侧和对照侧L4~6背根神经节,采用Western blotting法检测背根神经节内CollagenⅠ、TGF-β1、p-ERK、p-JNK、p-p38蛋白。结果与对照侧相比,造模8周时卡压侧腓肠肌内CSA降低、CVF升高(P均<0.05)。与对照侧相比,卡压侧背根神经节内CollagenⅠ、TGF-β1、p-ERK蛋白表达升高(P<0.05),但p-JNK、p-p38蛋白表达降低(P<0.05)。结论大鼠坐骨神经慢性卡压损伤可以引起背根神经节内CollagenⅠ上调,可能与背根神经元内TGF-β1表达升高有关,推测ERK信号通路激活参与了背根神经节内细胞外基质沉积和纤维化。Objective To investigate whether chronic sciatic nerve compression induces fibrosis in doral root gonglion(DRG)and to investigate mechanism of fibrosis change in the doral root gonglion.Methods Thirty male Sprague-Dawley rats(7-8 w)were subjected to sciatic nerve compression operation according to the method described by Mackinnon.The right sciatic nerve was operated to serve as ipsilateral side,while the left sciatic nerves were exposed as a comparative control.At 3 or 8 weeks after the establishment of the animal model,the ipsilateral and contralateral gastrocnemius muscles were performed via Masson's trichrome staining.Images were randomly selected to measure the average muscle fiber cross-sectional area(CSA)and collagen volume fraction(CVF).L4/L5/L6 ipsilateral and contralateral dorsal root ganglions were collected at 3 weeks after modeling.Western blotting was performed to determine the expression levels of collagenⅠ,transforming growth factor-β1(TGF-β1),phosphorylated extracellular signal-regulated kinase(p-ERK),phosphorylated c-Jun N-terminal kinase(p-JNK),and phosphorylated p38 mitogen-activated protein kinase(p-p38)in ipsilateral and contralateral dorsal root ganglions.Results Chronic sciatic nerve compression in the model group was associated with lower level of gastrocnemius muscle fiber CSA and higher percentage of gastrocnemius muscle fiber CVF at 8 weeks after surgery(P<0.05).Compared with the contralateral gastrocnemius muscle,the protein expression levels of collagenⅠ,TGF-β1,and p-ERK increased significantly in the ipsilateral DRGs(all P<0.05),while the levels of p-JNK and p-p38 decreased(both P<0.05).Conclusion Chronic sciatic nerve compression induces increases in collagenⅠin the DRGs,which may be associated with the increase of TGF-β1 protein expression in DRGs,and ERK signaling pathway may be involved in extracellular matrix deposition and fibrosis in DRGs.
