摘  要:目的:研究MAML1在肝内胆管细胞癌(intrahepatic cholangiocarcinoma,ICC)组织中的表达,阐明其对ICC早期诊断的作用。方法:在癌症基因图集数据库(the cancer genome atlas,TCGA)中分析MAML1基因,运用免疫组织化学(immunohistochemistry,IHC)技术分析MAML1在ICC组织及正常胆管组织的表达差异,阐述MAML1基因在ICC发生、发展进程的作用。结果:ICC组织中MAML1的mRNA表达量上升(P<0.05);在病理分级低的ICC组织中,MAML1的蛋白表达量高,相反,在病理分级高的组织中,MAMAL1蛋白表达量低。结论:MAML1在ICC中可能起着促癌基因的作用,临床上根据ICC组织中MAML1表达量的差异,可辅助鉴别ICC病理分级,能为临床工作者提供针对ICC患者制定个性化的治疗方案提供重要依据。Objective:To illuminate the effect of mastermind like transcriptional coactivator 1(MAML1)in diagnosis of intrahepatic cholangiocarcinoma(ICC)by studying the clinical expression of MAML1 in ICC tissue and benign tissue.Methods:MAML1 expression in ICC tissue and benign tissue was measured by using the Cancer Genome Atlas(TCGA).Further statistical analysis of immunohistochemistry(IHC)was performed to illuminate the role of MAML1 on ICC occurrence and progression.Results:Compared with the benign tissue,TCGA results showed that MAML1 expression was significantly up-regulated in the ICC tissue(ICC 1058.12±278.82vsbenign 480.37±86.11,P<0.01).In addition,IHC results showed MAML1 expression was significantly associated with low pathological grade(P<0.05).Conclusion:MAML1 may play a part as a carcinogenicgene in tumorigenesis.We can distinguish primary ICC samples,non-malignant samples and ICC samples with different pathological grade according to its different clinical expression.
