摘  要:目的探讨IL-13、COX2和PI3K/Akt信号通路与结肠癌侵袭转移的关系.方法以体外培养结肠癌细胞株HCT-8为研究对象,分为3组,A组经IL-13单独作用,B组无IL-13或IFN-γ作用,C组经IFN-γ单独作用.RT-PCR法检测3组细胞IL-13、COX2、PIK3CA、Akt1、磷酸化蛋白激酶B(p-AKT)的蛋白表达水平,并通过Transwell小室侵袭实验观察比较3组细胞的侵袭能力以及划痕实验观察比较3组细胞的48 h转移能力状况.结果与B组比较,A组IL-13处理后细胞COX2的mRNA相对表达量以及IL-13、COX2、p-AKT蛋白表达水平均提高,C组IFN-γ处理后细胞IL-13、COX2的mRNA相对表达量以及IL-13、COX2、p-AKT蛋白表达水平均降低(P<0.05).与C组比较,A组IL-13处理后细胞IL-13、COX2的mRNA相对表达量以及IL-13、COX2、p-AKT蛋白表达水平均较高(P<0.05).与同组处理前比较,A组IL-13处理后细胞COX2的mRNA相对表达量以及IL-13、COX2、p-AKT蛋白表达水平均提高,C组IFN-γ处理后细胞IL-13、COX2的mRNA相对表达量以及IL-13、COX2、p-AKT蛋白表达水平均降低(P<0.05).3组中,处理后,A组48 h细胞迁移距离最长且Transwell穿膜细胞数最多,其次为B组增加,再次为C组,差异有统计学意义(P<0.05).A组IL-13作用后细胞48 h细胞迁移距离和Transwell穿膜细胞数增加,C组IFN-γ处理后48 h细胞迁移距离和Transwell穿膜细胞数减少(P<0.05).结论IL-13、COX2和PI3K/Akt信号通路均与结肠癌侵袭转移相关,其中可能机制为IL-13调控COX2和PI3K/Akt信号通路促进结肠癌侵袭转移.Objective To investigate the correlation between IL-13,COX2 and PI3K/Akt signaling pathways and the invasion and metastasis of colon cancer.Methods Colon cancer cell line HCT-8 was cultured in vitro.The mRNA expression of IL-13,COX2,PIK3CA and Akt1 was assess by RT-PCR in co-cultured HCT-8 cells with IL-13 only(Group A),without IL-13 or IFN-γ(Group B)and IFN-γonly(Group C).The protein expression of IL-13,COX2,PIK3CA,Akt1 and phosphorylated protein kinase B(p-AKT)of the 3 groups was assessed by Western blot.The invasive ability and 48-hour metastasis ability were compared by Transwell cell invasion test and scratch test.Results Compared with those of Group B after treatment,the mRNA expression of COX-2 and protein expression levels of IL-13,COX-2,p-AKT in Group A after IL-13 treatment were significantly increased,while the mRNA expres-sion of IL-13,COX-2 and protein expression levels of IL-13,COX-2,p-AKT in Group C after IFN-γtreatment were significantly reduced(P<0.05).Compared with those of Group C after treatment,the mRNA expression of IL-13,COX-2 and protein expression levels of IL-13,COX-2,p-AKT in Group A after IL-13 treatment were significantly increased(P<0.05).Compared with those of the same group before treatment,the mRNA expression of COX-2 and protein expression levels of IL-13,COX-2,p-AKT in Group A after IL-13 treatment were increased,while the mR-NA expression of IL-13,COX-2 and protein expression levels of IL-13,COX-2,p-AKT in Group C after IFN-γtreatment were significantly reduced(P<0.05).Among the three groups,48 hours after treatment,Group A had the lon-gest cell migration distance and the largest number of transmembrane cells,followed by the Group B and Group C,and the difference was statistically significant(P<0.05).In Group A,the cell migration distance and the number of transmem-brane cells significantly increased 48 hours after IL-13 treatment;while in Group C the cell migration distance and the number of transmembrane cells were significantly reduced 48 hours after IFN-γtreatment(P<0.
