摘  要:目的探讨VX2骨肿瘤模型兔血清干扰素(interferon, IFN)-γ和白细胞介素(interleukin, IL)-4水平变化及意义。方法 40只日本大耳白兔随机分为骨肿瘤模型组和对照组各20只,其中骨肿瘤模型组采用VX2肿瘤组织块种植于兔胫骨建立兔VX2骨肿瘤模型,对照组采取同样手术操作但不植入VX2肿瘤。植瘤后第7天采用自主设计通道内置式骨肿瘤病理组织取出装置验证成瘤率;植瘤后第7、14天采用ELISA法检测2组兔血清IFN-γ和IL-4水平,处死兔后取胫骨组织行组织病理学检查。结果 37只实验兔纳入最终实验,骨肿瘤模型组18只,对照组19只。经穿刺活检装置证实成瘤率100%,穿刺成功率100%。骨肿瘤模型组植瘤第7天肿瘤组织在胫骨髓腔内生长,骨皮质未见肿瘤细胞;植瘤第14天肿瘤组织于胫骨髓腔内生长面积增大,骨皮质发现肿瘤细胞,5只实验兔穿刺孔处软组织触及肿块并发现肿瘤细胞。骨肿瘤模型组植瘤第14天血清IFN-γ水平[(697.3±39.1)ng/L]低于第7天[(734.7±27.6)ng/L](P<0.05),IL-4水平[(207.2±18.6)ng/L]高于第7天[(176.5±14.2)ng/L](P<0.05);对照组植瘤第14天血清IFN-γ、IL-4[(749.4±25.6)ng/L、(171.4±12.7)ng/L]水平与第7天[(747.7±27.8)ng/L、(169.5±12.8)ng/L]比较差异无统计学意义(P>0.05)。骨肿瘤模型组植瘤第7天血清IFN-γ、IL-4水平与对照组比较差异无统计学意义(P>0.05),植瘤第14天血清IFN-γ水平低于对照组(P<0.05),IL-4水平高于对照组(P<0.05)。结论 VX2肿瘤种植于兔胫骨后,随着肿瘤的生长机体抗肿瘤免疫反应受到抑制,表现为IFN-γ水平下降,IL-4水平上升。Objective To investigate the changes and significances of serum interferon(IFN)-γ and interleukin(IL)-4 in VX2 bone tumor rabbit models. Methods Forty Japanese white rabbits were randomly divided into model group and control group, with 20 rabbits in each group. VX2 tumor tissue was implanted in the tibia to establish VX2 bone tumor models in model group, and control group was performed the same operation procedures but without VX2 tumor tissue implantation. On the 7 th day after tumor implantation, the self-designed bone tumor pathological tissue extraction device was used to verify the tumor formation rate. The peripheral blood of rabbits was collected on the 7 th and 14 th days after tumor implantation, and the levels of IFN-γ and IL-4 in peripheral blood were detected by ELISA method. After the rabbits were killed, the tibial tissues were taken for pathological examination. Results Totally 37 experimental rabbits were included in the final experiment, 18 in model group and 19 in control group. The puncture biopsy device confirmed that the tumor formation rate was 100%, and the puncture success rate was 100%. In model group, the tumor tissue grew in the tibial marrow cavity on the 7 th day after tumor implantation, and no tumor cells were found in the bone cortex;the tumor tissue increased in the tibial marrow cavity on the 14 th day after tumor implantation, tumor cells were found in the bone cortex, and tumor cells were found in the soft tissue of the puncture hole in 5 rabbits. In model group, the level of IFN-γ was lower and the level of IL-4 was higher on the 14 th day after tumor implantation((697.3±39.1),(207.2±18.6)ng/L) than that on the 7 th day after tumor implantation((734.7±27.6),(176.5±14.2)ng/L)(P<0.05). In control group, the levels of IFN-γ and IL-4 showed no significant differences on the 14 th day((749.4±25.6),(171.4±12.7)ng/L)compared with those on the 7 th day((747.7±27.8),(169.5±12.8)ng/L)(P>0.05).The levels of IFN-γand IL-4 showed no significant differences between model group
