摘 要:目的:分析c-Met、miRNA-1在宫颈癌组织中的表达水平变化,以及其与癌组织病理特征之间的相关性,探究miRNA-1和c-Met与宫颈癌生物学行为的关系。方法:收集内蒙古自治区人民医院2015年3月至2017年3月收治的50例宫颈癌患者的肿瘤组织及癌旁组织,采用免疫组织化学法检测癌组织及癌旁组织中c-Met表达;采用RT-PCR检测癌组织及癌旁组织中miRNA-1的相对表达量;采用配对t检验对比分析癌组织与癌旁组织中c-Met、miRNA-1表达;应用Spearson线性相关检验分析c-Met、miRNA-1表达水平与宫颈癌患者临床病理特征间的相关性;应用Kaplan-Meier和Log-rank检验统计分析c-Met、miRNA-1表达对宫颈癌患者生存情况的影响。结果:宫颈癌组织中c-Met表达高于癌旁组织,24个月随访结果显示,c-Met表达越低,宫颈癌患者生存率越高。癌组织中miRNA-1表达水平显著低于癌旁组织,为癌旁组织的(0.41±0.07)倍,差异有统计学意义(P<0.01);miRNA-1表达水平越低,患者生存率越低。结论:在宫颈癌组织中miRNA-1表达下调,c-Met表达上调,其表达与宫颈癌发生发展、预后有关,提示miRNA-1和c-Met可作为宫颈癌诊断和预后的重要生物学指标。Objective:To analyze the changes of c-met and miRNA-1 expression in cervical cancer tissues and their correlation with the pathological features of cancer tissues,to explore the relationship between miRNA-1,c-Met and the biological behaviors of cervical cancer.Methods:Tumor tissues and the adjacent tissues were collected from 50 cases of cervical cancer patients admitted to People’s Hospital of Inner Mongolia Autonomous Region from March 2015 to March 2017.c-Met expression was examined using immunohistochemical method;the relative expression level of miRNA-1 was detected using RT-PCR;the comparative analyses of c-Met and miRNA-1 expression between tumor tissues and the adjacent tissues were conducted using paired t test;the correlation between c-Met and miRNA-1 expression and cervical cancer patients’clinical pathological features was analyzed using Spearson linear correlation test;the impact of c-Met and miRNA-1 expression on cervical cancer patients’survival was analyzed using the Kaplan-Meier and Log-rank tests.Results:In cervical cancer tissues,c-Met had a higher expression.The 24-month follow-up showed that the lower the c-Met expression was,the higher the cervical cancer patients’survival rate was.The miRNA-1 had a(0.41±0.07)-time lower expression in cancer tissues than in the adjacent tissues,indicating a statistical significance(P<0.01);The lower the miRNA expression was,the lower the patient’s survival rate was.Conclusion:The miRNA-1 expression is down-regulated while The c-Met expression is up-regulatedin in cervical cancer tissues,which is related to development and prognosis of cervical cancer.suggesting that miRNA-1 and c-Met can be used as important biological indicators for the diagnosis and prognosis of cervical cancer.
