X染色体耦联锌指蛋白表达与胃癌组织病理及预后的相关性

摘　　要：目的观察X染色体耦联锌指蛋白(ZFX)在胃癌组织中的表达及与胃癌组织病理、预后的相关性。方法回顾性分析2018年1月至2019年6月秦皇岛市第一医院收治的112例胃癌患者的病例资料,对手术切除的胃癌组织标本予以观察,癌旁组织标本作为对照,检测胃癌组织与癌旁组织ZFX mRNA表达情况,分析ZFX蛋白与临床病理参数及患者预后的相关性。结果经过逆转录聚合酶链反应(RT-PCR)检测,发现ZFX mRNA蛋白在胃癌组织中的表达量为(6.38±2.31)mRNA,显著高于癌旁组织的(1.52±0.75)mRNA,差异具有统计学意义(t=11.204,P<0.05);免疫组织化学(IHC)结果显示,ZFX蛋白表达位于细胞核,其表达阳性率达到76.77%,高于癌旁组织的18.75%,差异具有统计学意义(χ^2=47.214,P<0.05);Logistic回归分析显示,胃癌组织中ZFX蛋白表达与肿瘤分化程度、淋巴结转移以及浸润深度等因素有关,差异具有统计学意义(t=6.903,4.842,5.514,P<0.05);ZFX蛋白高表达组、低表达组患者总生存期分别为(35.28±2.14)个月、(54.29±4.16)个月,差异具有统计学意义(t=12.192,P<0.05)。结论胃癌组织中ZFX蛋白呈现出异常高表达,与患者病理参数及预后相关,该指标可用于对患者潜在预后的评估。Objective To observe the expression of X-linked zinc finger protein(ZFX)in gastric cancer and its correlation with the histopathology and prognosis of gastric cancer.Methods The clinical data of 112 patients with gastric cancer who were treated in Qinhuangdao First Hospital from January 2018 to June 2019 were analyzed retrospectively.The samples of surgically resected gastric cancer tissues were observed.The samples of paracancerous tissues were used as controls.The expression of ZFX mRNA in gastric cancer and paracancerous tissues were detected,and the correlation between ZFX protein and clinical pathological parameters and prognosis of patients was analyzed.Results The results of reverse transcription polymerase chain reaction(RT-PCR)showed that the expression of ZFX mRNA protein in gastric cancer tissues was(6.38±2.31)mRNA,which was significantly higher than that of paracancerous tissues(1.52±0.75)mRNA,and the difference was statistically significant(t=11.204,P<0.05).The results of immunohistochemistry(IHC)showed that the expression of ZFX protein was located in the nucleus,and its positive expression rate reached 76.77%,which was higher than that of paracancerous tissues(18.75%),the difference was statistically significant(χ^2=47.214,P<0.05).Logistic regression analysis showed that the expression of ZFX protein in gastric cancer tissue was related to factors such as tumor differentiation,lymph node metastasis and depth of invasion,and the differences were statistically significant(t=6.903,4.842,5.514,P<0.05).The overall survival of patients in the ZFX protein high expression group and low expression group were(35.28±2.14)months and(54.29±4.16)months,respectively,and the difference was statistically significant(t=12.192,P<0.05).Conclusion The expression of ZFX protein in gastric cancer is abnormally high,which is related to the pathology and prognosis,and can be used to evaluate the potential prognosis of patients.
