摘 要:目的探讨2型糖尿病(type 2 diabetes mellitus,T2DM)遗传易感性与LEPR基因rs1892534、rs2211651、rs1805096位点多态性及环境因素的交互作用。方法病例组来源于内蒙古医科大学附属医院、中医医院及国际蒙医医院的203例2型糖尿病患者,对照组为203例健康体检人群。多重高温连接酶检测反应(iMLDR)检测LEPR基因SNP位点多态性,广义多因子降维法(GMDR)分析LEPR基因与环境交互作用。结果LEPR基因rs1805096、rs1892534、rs2211651基因型频率分布在病例组和对照组之间的差异有统计学意义(P<0.05),单体型GCG在两组间差异有统计学意义;交互作用分析结果显示,一阶交互最优模型是rs1805096、高脂饮食(P<0.05);二阶模型rs1805096、rs1892534及高血压疾病史模型和rs1805096、rs2211651及高脂饮食模型均存在交互作用(P<0.05)。结论呼和浩特地区汉族人群LEPR基因rs1805096、rs1892534、rs2211651单核苷酸多态性与T2DM易感性相关,GCG是T2DM的易感单体型,LEPR基因-环境的交互作用可能会增加T2DM的发病风险。Objective To explore the interaction between genetic susceptibility of T2 DM and LEPR geners1892534,rs2211651,rs1805096 polymorphism and environmental factors.MethodsThe case group wasderived from 203 patients with type 2 diabetes in the Affiliated Hospital of Inner Mongolia Medical University,Chinese Medicine Hospital and International Mongolian Medical Hospital,and the control group was 203 healthypeople.Multiple high temperature ligase detection reaction(i MLDR)was used to detect LEPR gene SNPpolymorphism.Generalized multi-factor dimensionality reduction(GMDR)analysis of the interaction between LEPRgene and environment.ResultsThe difference in genotype frequency distribution of the LEPR gene rs1805096,rs1892534,rs2211651 between the case group and the control group was statistically significant(P<0.05),andthe haplotype GCG was statistically significant between the two groups;Interaction analysis results show that thefirst-order interaction optimal model rs1805096,high-fat diet(P<0.05);the second-order models rs1805096,rs1892534 and the history of hypertension disease model and rs1805096,rs2211651 and high-fat diet model haveinteractions(P<0.05).ConclusionThe LEPR gene rs1805096,rs1892534,and rs2211651 single nucleotidepolymorphisms in the Han population in Hohhot are associated with T2 DM susceptibility.GCG is a susceptiblehaplotype of type 2 diabetes,and the LEPR gene-environment interaction may increase the risk of type 2 diabetes.
