摘 要:优化超声波辅助提取凹纹胡蜂蜂房总黄酮(Vespa auraria Smith nidus vespae totalavonoids,VASNVTF)提取工艺,并测定其总黄酮的抗氧化性,为VASNVTF的开发利用提供技术支持。在单因素试验的基础上,以总黄酮提取率为评价指标,通过L9(34)试验优化其总黄酮的提取工艺条件,并通过测定VASNVTF对Fe^3+还原能力和对羟基自由基(·OH)、1,1-苯基-2-苦肼基自由基(DPPH·)的清除率评价抗氧化性。结果表明:最佳提取工艺为乙醇体积分数55%、超声温度50℃、超声时间80 min、液料比70 mL/g。在此条件下总黄酮平均提取率为2.32%,RSD值为0.43%。VASNVTF质量浓度为0.0186 g/L时,使Fe3+还原产生的吸光值可达到1.03,对·OH和DPPH·的清除率可分别达到96.25%和99.40%。试验表明,VASNVTF有较强抗氧化性,可作为一种抗氧化剂进行利用。Ultrasonic assisted extraction of total favonoids from Vespa araria Smith nidus vespae(VASNVTF),and antioxidant activity of total favonoids in Vespa araria Smith nidus vespae was evaluated to provided technique support for the development and utilization of VASNVTF.Optimization of extraction parameters for improved extraction efficiency of total flavonoids was done using single factor experiment and an orthogonal array design L9(34).Meanwhile,the antioxidant activity of VASNVTF was evaluated by the reduction of Fe^3+and scavenging of hydroxyl radicals(·OH)and 1,1-phenyl-2-picryazide radicals(DPPH·).The optimum extraction parameters were determined as follows:ethanol volume fraction was 55%,ultrasonic temperature was 50℃,ultrasonic time was 80 min,liquid material ratio was 70 mL/g.Under these conditions,the average extraction yield of total flavonoids was 2.32%,and RSD value was 0.43%.When the concentration of VASNVTF was 0.0186 g/L,the absorbance value of Fe3+reduction could reach 1.03,and the percentage scavenging of·OH and DPPH·were 96.25%and 99.40%,respectively.The experiment indicated that the VASNVTF had strong antioxidant activity in vitro,it can be used as a natural antioxidant for development and utilization.
