摘　　要：目的探讨粉防己碱对SMMC-7721细胞体外生长和体内成瘤的影响。方法CCK8法检测细胞活力;Hoechst染色检测细胞凋亡;Transwell检测细胞侵袭;划痕实验检测细胞迁移;Western印迹检测Ki67、Caspase-3、MMP-9、VEGF、c-Fos、c-Myc表达和ERK1/2磷酸化水平;建立移植瘤模型,检测肿瘤重量,免疫组化检测Ki67和VEGF表达。结果体外实验:粉防己碱抑制细胞活力、侵袭和迁移(P<0.05),促进细胞凋亡(P<0.05),下调Ki67、Caspase-3、MMP-9、VEGF、c-Fos、c-Myc表达(P<0.05),降低ERK1/2磷酸化水平(P<0.05)。体内实验:粉防己碱降低肿瘤重量(P<0.05),下调Ki67、VEGF表达(P<0.05)。结论粉防己碱抑制ERK1/2磷酸化,从而抑制SMMC-7721细胞体外生长和体内成瘤。Objective To investigate the effect of tetrandrine on SMMC-7721 cell growth in vitro and tumor formation in vivo.Methods The cell activity was detected by CCK8 method,apop-tosis by Hoechst staining,cell invasion by Transwell,cell migration by scratch test and the expres-sion levels of Ki67,caspase-3,matrix metallopeptidase 9(MMP-9),VEGF,c-Fos,c-Myc and extracellular signal.regulated protein kinase 1/2(ERK1/2)phosphorylation by Western blotting.A xenograft tumor model was established to detect tumor weight,and expression levels of Ki67 and VEGF were immunohistochemically detected.Results In vitro experiment showed that tetrandrine inhibited cell activity,invasion and migration(P<0.05),promoted apoptosis(P<0.05),down-regulated the expression levels of Ki67,caspase-3,MMP-9,VEGF,c-Fos,c-Myc expres-sion(P<0.05),and decreased ERK1/2 phosphorylation(P<0.05).In vivo experiment re-vealed that tetrandrine decreased tumor weight(P<0.05),and down-regulated Ki67 and VEGF expression(P<0.05).Conclusion Tetrandrine inhibits ERK1/2 phosphorylation,thereby inhib-iting SMMC-7721 cell growth in vitro and tumor formation in vivo.
