摘 要:目的探讨Neurexin、5-羟色胺受体4(5-HT4R)蛋白在小儿先天性巨结肠中的表达及意义。方法选取2017年1月至2018年2月青海省妇女儿童医院保存的先天性巨结肠标本18例作为观察组,同时选取正常肠管12例作为对照组,采用免疫组化染色法检测Neurexin蛋白、5-HT4R蛋白表达水平。结果观察组各段Neurexin蛋白和5-HT4R蛋白表达水平明显低于对照组,差异有统计学意义(P<0.05)。观察组狭窄段Neurexin和5-HT4R蛋白表达均明显低于扩张段和移行段,差异有统计学意义(P<0.05);观察组移行段Neurexin蛋白表达水平明显低于扩张段(P<0.05);观察组移行段和扩张段5-HT4R蛋白表达比较无统计学意义(P>0.05)。观察组患者扩张段、移行段和狭窄段Neurexin蛋白表达水平与5-HT4R蛋白表达水平呈正相关(r=0.797、0.686、0.579,P<0.05)。结论Neurexin蛋白、5-HT4R蛋白在小儿先天性巨结肠中的表达水平明显降低,两者间有一定相关性,可能在疾病发生发展中有重要作用。Objective To investigate the expression and significance of 5,5-hydroxytryptamine serotonin receptor 4(5-HT4R) protein in children with Hirschsprung’s disease.Methods Eighteen cases of congenital megacolon specimens preserved in our hospital from January 2017 to February 2018 were selected as observation group,and 12 normal intestinal tubes were selected as control group.Neurenxin protein and 5-HT4 R protein were detected by immunohistochemical staining.The expression level.Results The expression levels of Neurexin protein and 5-HT4 R protein in the observation group were significantly lower than those in the control group(P<0.05).The expression of Neurexin and 5-HTR protein in the stenosis group were significantly lower than the expansion segment and transition segment,the difference was statistically significant(P<0.05);the expression level of Neurexin protein in the observation group was significantly lower than the expansion segment(P<0.05).There was no significant difference in the expression of 5-HT4 R protein between the transition group and the dilated segment of the observation group(P>0.05).The expression level of Neurexin protein in the expansion,transition and stenosis segments was positively correlated with the expression of 5-HT4 R protein(r=0.797,0.686,0.579,P<0.05).Conclusion The expression levels of Neurexin and 5-HT4 R proteins in children with Hirschsprung’s disease are significantly decreased.There is a certain correlation between them,which may play an important role in the development of diseases.
