摘 要:卵巢癌在妇科三大恶性肿瘤中的死亡率位居首位。随着卵巢癌手术技术的不断地进步,以铂为基础的联合化疗也取得了明显进展。然而近30年来,晚期卵巢癌患者的5年生存率并没有明显提高。已经被广泛应用多年的药物评估模型——肿瘤细胞系模型,由于其肿瘤生物学特征和原发肿瘤相去甚远,在预测临床药效指导治疗上存在一定局限性。由此,人源性异种移植瘤(patient-derived xenograft,PDX)模型应运而生,由于其能够保留供体肿瘤的主要组织学和遗传学特征,在肿瘤研究领域中已显示出一定的优势。本文将就卵巢PDX模型相较于传统模型的优势,以及其在卵巢癌药物测试、药物研发及临床上的应用和局限性进行综述。Ovarian cancer ranks first among three major gynecological malignancies.With surgical techniques for ovarian cancer having continued to advance,significant progress has been made in platinum-based combination chemotherapy.However,the five-year survival rate of patients with advanced ovarian cancer has not improved significantly in the past 30 years.The tumor cell line model,a drug evaluation model that has been widely used for many years,has a certain limitation in predicting clinical efficacy and guiding treatment due to its tumor biological characteristics far from the primary tumor.As a result,a patient-derived xenograft(PDX)model emerged at the historic moment.It can retain the main histological and genetic characteristics of the donor tumor,it has shown certain advantages in the field of tumor research.This review will discuss the advantages of the ovarian PDX model with traditional models,and the application and limitations of the ovarian PDX model in ovarian cancer drug testing,drug development and clinical application.
