山竹果皮提取物抑制非小细胞肺癌细胞增殖、迁移和侵袭的机制研究

摘　　要：目的探讨山竹果皮提取物是否通过调控SPOCK1表达影响非小细胞肺癌细胞的增殖、迁移和侵袭。方法用不同终浓度(100μmol/L、200μmol/L和400μmol/L)的山竹果皮提取物处理A549细胞24 h,CCK-8法检测细胞增殖活力,Transwell法检测细胞的迁移和侵袭能力,qRT-PCR和Western blot检测SPOCK1 mRNA和SPOCK1蛋白的表达。建立抑制SPOCK1表达A549细胞株,观察其对A549细胞增殖、侵袭和迁移能力的影响。建立过表达SPOCK1的A549细胞株,然后再给予山竹果皮提取物(400μmol/L)处理24 h,观察其对A549细胞增殖、侵袭和迁移能力的影响。结果山竹果皮提取物呈剂量依赖性地抑制A549细胞的增殖、迁移和侵袭,抑制SPOCK1的表达(P<0.05))。抑制SPOCK1表达可抑制非小细胞肺癌A549细胞增殖、迁移和侵袭(P<0.05)。过表达SPOCK1可逆转山竹果皮提取物对A549细胞增殖、迁移侵袭的抑制作用(P<0.05)。结论山竹果皮提取物通过下调SPOCK1从而抑制非小细胞肺癌细胞的增殖、迁移和侵袭。Objective To investigate whether mangosteen peel extract can affect the proliferation,migration and invasion of non-small cell lung cancer cells by down-regulating the expression levels of SPOCK1.Methods A549 cells were treated with the mangosteen peel extract at different final concentrations(100μmol/L,200μmol/L and 400μmol/L)for 24h.And CCK8,Transwell assay,Western blot and qRTPCR were used to detect the cell proliferation activity,the migration and invasion ability of cells and the expression of SPOCK1 mRNA and SPOCK1 protein,respectively.The A549 cell lines with overexpression of SPOCK1 were established to observe its effects on the proliferation,migration and invasion of A549 cells.And A549 cell lines over expressing SPOCK1 was established and were treated with mangosteen peel extracts at 400μmol/L for 24 hours,then its effects on the proliferation,migration and invasion of A549 cells were observed.Results The mangosteen peel extract inhibited the proliferation,migration,invasion of A549 cells and the expression of SPOCK1 in a dose-dependent manner(P<0.05).The inhibition of the expression of SPOCK1 could inhibit the proliferation,migration and invasion of non-small cell lung cancer cells(P<0.05).Overexpression of SPOCK1 reversed the inhibitory effects of mangosteen peel extract on the proliferation,migration and invasion of A549 cells(P<0.05).Conclusion The mangosteen peel extract can inhibit the proliferation,migration and invasion of non-small cell lung cancer cells by down-regulating the expression of SPOCK1.
