摘  要:目的:研究谷红注射液(GHI)对大鼠骨折愈合的促进作用,以及对成纤维细胞生长因子2(FGF-2)表达的影响。方法:选取雄性SD大鼠72只,随机分为3组,分别为对照组(生理盐水)、GHI组[10 ml/(kg·d)]、阳性药组[复方骨肽注射液,5 ml/(kg·d)],每组24只。各组大鼠均在麻醉状态下建立开放式截骨骨折模型,第2、4、6周时拍射X线进行影像学评估,HE染色观察骨折病理变化,同时进行离体胫骨骨矿物质密度(BMD)、血清碱性磷酸酶(ALP)及成纤维细胞生长因子2(FGF-2)测定,采用免疫组化法检测软骨中FGF-2蛋白表达。结果:与对照组相比,建模后第2、4周GHI组、阳性药组X线骨组织愈合评分均明显提高(P<0.05);第2周时,阳性药组和GHI组HE切片中软骨细胞数量多于对照组。第4周时,对照组骨折端仍以软骨组织为主,阳性药组和GHI组的软骨组织及纤维组织逐渐被不成熟骨组织替代;治疗后第2、4、6周均显示,GHI组、阳性药组BMD值高于对照组(P<0.05),但GHI组与阳性药组差异无统计学意义;建模治疗后第2周,GHI组、阳性药组血清ALP水平均高于对照组(P<0.05),而第2、4、6周GHI组、阳性药组血清FGF-2水平均高于对照组(P<0.01);在第2、4周,各组间FGF-2蛋白表达均有显著性差异,而第6周时,各组间FGF-2蛋白表达差异无统计学意义。结论:谷红素注射液对促进骨折愈合有明显效果,其作用可能与促进FGF-2表达有关。Objective:To study the effect of GHI on fracture healing and the expression of fibroblast growth factor-2(FGF-2)in rats.Methods:72 male SD rats were randomly divided into three groups:control group(normal saline),GHI group[210 mg/(kg·d)]and positive drug group[compound osteopeptide injection,5 mg/(kg·d)],24 rats in each group.Rats in each group were established open fracture models under anesthesia.Radiographic evaluation was performed at 2nd,4th,6th week.The pathological changes of fracture were observed by HE.Bone mineral density(BMD),serum alkaline phosphatase(ALP)and fibroblast growth factor-2(FGF-2)were measured in vitro.Immunohistochemistry was used to detect the expression of FGF-2 protein in cartilage.Results:Compared with control group,the X-ray bone healing score of GHI group and positive drug group were increased significantly at 2nd and 4th week after modeling(P<0.05).At 2nd week,the number of chondrocytes in the HE slice of GHI group and positive drug group was more than that of the control group.At 4th week,cartilage tissue was still the main fracture site in the control group,and cartilage tissue and fibrous tissue in GHI group and positive drug group were gradually replaced by immature bone tissue;BMD values of GHI group and positive drug group were higher than those of control group at the 2nd,4th and 6th week(P<0.05),but there was no significant difference between GHI group and positive drug group;the serum ALP levels in GHI group and positive drug group were higher than those in the control group at the 2nd week(P<0.05),while the serum FGF-2 levels in GHI group and positive drug group were higher than those in the control group at the 2nd,4th and 6th week(P<0.01)At the 2nd and 4th week,the expression of FGF-2 protein was significant differences among the groups.Conclusion:GHI has obvious effect on promoting fracture healing,and its effect may be related to the promotion of FGF-2 expression.
