摘 要:目的:研究miR-9在急性缺血性脑卒中(AIS)患者血清中表达水平,并探讨miR-9表达与AIS患者血清炎症关系。方法:收集2017年6月~2019年6月在唐山市人民医院住院治疗的51例AIS患者血清,同时收集同期于我院体检的23名健康志愿者血清,RT-qPCR检测miR-9表达水平,ELISA测定血清炎症因子,双荧光素酶基因报告系统验证miR-9的靶基因,Western blot检测蛋白表达水平。结果:miR-9在AIS患者血清中相对表达升高,并且其表达水平与AIS患者NIHSS评分(r=0.674,P<0.001)、脑梗死体积(r=0.544,P<0.001)、血清IL-6(r=0.558,P<0.001)、IL-8(r=0.581,P<0.001)、TNF-α(r=0.626,P<0.001)及IL-1β(r=0.668,P<0.001)含量均呈正相关。miR-9-mimic可显著降低THP-1细胞和健康志愿者PBMC中SIRT1蛋白表达水平(P<0.001);miR-9-inhibitor可以显著升高THP-1细胞和健康志愿者PBMC中SIRT1蛋白表达水平(P<0.001)。结论:miR-9在AIS患者血清中高表达,高表达的miR-9与AIS患者病情及血清炎症呈正相关,可能与miR-9抑制PBMC SIRT1表达有关。Objective:To investigate expression of miR-9 in serum of patients with actue ischemic stroke(AIS),and to explore the correlation between miR-9 expression and inflammation in serum of patients with AIS.Methods:Serum of 51 patients with AIS and serum of 23 healthy volunteers in Tangshan People′s Hospital from June 2017 to June 2019 were collected.Expression of miR-9 was detected by RT-qPCR,level of inflammationary factors were measured by ELISA,target gene of miR-9 was verified by a dual luciferase gene reporter system,and protein expression level was detected by Western blot.Results:Relative expression of miR-9 in serum of 51 patients with AIS was increased,and expreesion of miR-9 was positively related to NIHSS score(r=0.674,P<0.001),cerebral infarction volume(r=0.544,P<0.001),IL-6(r=0.558,P<0.001),IL-8(r=0.581,P<0.001),TNF-α(r=0.626,P<0.001)and IL-1β(r=0.668,P<0.001).miR-9-mimic could significantly reduce expression level of SIRT1 protein in THP-1 cells and healthy human PBMC(P<0.001),miR-9-inhibitor could significantly increase expression level of SIRT1 protein in THP-1 cells and healthy human PBMC(P<0.001).Conclusion:miR-9 is highly expressed in serum of patients with AIS,and high expression of miR-9 is positively correlated with condition and serum inflammation in patients with AIS.This may be related to that miR-9 can inhibited expression of SIRT1 in PBMC.
